Human T-cell leukemia viruses type 1 and 2 (HTLV-1 and HTLV-2) share a common genetic organization, expression strategy and ability to infect and immortalize T-cells in vitro; however, HTLV-1 and HTLV-2 are strikingly different in terms of clinical impact. HTLV-1 is recognized as the aetiological agent of adult T-cell leukemia/lymphoma (ATLL), and HTLV-associated myeolopathy/tropical spastic paraparesis (HAM/TSP), in contrast, HTLV-2 does not cause hematologic disorders and is only sporadically associated with cases of subacute myelopathy. HTLV-1 and HTLV-2 also exhibit distinct cellular tropisms in vivo: HTLV-1 is mainly found in CD4+T lymphocytes, whereas CD8+T-cells are the preferred target for HTLV-2. The articles contributed in this Research Topic are covering all the different aspects that characterize HTLV-1 and HTLV-2, by highlighting differences in their biology that might provide clues to their distinct pathogenic properties.

The individual chapters represent the synopses of contributions which were presented at the Seventh International Symposium on ADP-Ribosylation Reactions, held in Vitznau, Switzer land, from September 23 to 27, 1984. This volume covers new devel opments in the field since the last meeting was held on this topic in 1982, in Tokyo. Therefore, the present text is not meant to form a comprehensive account of a specialized research area, but encompasses a collection of state-of-the-art reports from the vast majority of laboratories currently involved in ADP-ribosylation work. For the sake of rapid publication, the editorial policy was to ensure easy accessibility of information contained in individual articles rather than to provide elaborate cross references or reference to work published prior to 1982. However, a detailed subject index will help the reader find complementary information. The enzymes of ADP-ribose metabolism have not yet acquired universally acceptable trivial names and the Enzyme Commission has not yet defmitely decided on formal appellations. Consequently, a variety of names for the nuclear enzyme appear in this book, including nuc1ear(ADP-ribosyl)transferase, poly(ADP-ribose) polymerase, or synthetase or synthase. Hopefully, a common convention will soon be established. This book contains the proceedings of a conference devoted to the study of the structure and function of Terminal deoxynucleotidyl Transferase (TdT) and its utilization as biochemical marker in immuno biology and leukemia, held in Elba, Italy on May 28-31, 1981. The en zyme has been known to nucleic acid biochemists for more than 20 years and has proved to be an excellent tool for making deoxypolymers, label ing DNA fragments, and adding homopolymer tails to restriction endonu clease fragments from DNA. Since the discovery of its peculiar tissue distribution, normally restricted to the thymus and bone marrow, and of its abnormal occur rence in human leukemic lymphoblasts, TdT has become one of the most widely used markers in the study of lymphocyte differentiation and in the classification of hematopoietic neoplasia. The subject seemed most appropriate for a meeting where molecular and cellular biologists, immunologists and hematologists could convene for the first time to discuss both basic research and the clinical aspects of the problem. Among the goals achieved by this workshop was the sharing of in formation about the enzymology of TdT, biochemical and immunological methodology and the correlation of TdT with other markers in the diag nosis of leukemia. The remarkable accordance of results, presented here by five independent hematological institutions, from analysis of TdT in several thousands of leukemic patients, marks the importance of this enzyme as a diagnostic and prognostic tool in these diseases.
