None of these properties are obviously associated with surface enzymes. What are the functions of the cell surface that may require the presence of enzymes? From a purely hypothetical point of view, the cellular activities in which cell surface enzymes are potentially functional can be divided into two categories; first, those activities related to the production and maintenance of the cell surface structure itself, and second, those activities which are concerned with interactions between the cell and its environment. During growth of a cell, as it enlarges, new structural ma terial must be synthesized and assembled in the cell membrane. Although it is conceivable that new structural material is synthesized in the interior of the cell, it is difficult to visualize its complete synthesis and final assembly in the periphery of the cell, without the assistance of enzymes at the site. In the case of non-growing cells, although no net synthesis of new structural material is necessary, the turnover of the various components by a continuous degradation and resynthesis must be considered as a pos sibility.

Since the publication of my monograph "Die submikroskopische Struktur des Cytoplasm as" [Protoplasmatologia III A12 (1955)], science has increased our knowledge on this cell constituent in an amazing way. At that time, cytoplasm was still considered as an amorphous colloid with an amicroscopic structure capable of sol/gel transformations. Meanwhile, within the past 18 years, ultrastructure research and molecular biology have shown that it consists of structured organelles, thereby revolutionizing our concept of the cytoplasm. There is an ontogeny of these organelles which deals with their appearance, development, growth and eventual disappearance. Although the results of relevant studies are less spectacular at this time than those concerning nucleic acids, they will become more and more important in the near future, because they touch the basic question of how molecular morphogenesis is realized. Therefore, an attempt is made to portray the actual state of these problems from a biological point of view which involves a comparison of the established organelles. It is also possible that contractile vacuoles originate by some form of phase separation not yet understood and possibly involving the contractile properties of proteins, and' it is possible that this process continues to operate in the walls of the vacuoles or feeder canals. This secretory process may be the function of that part of the vacuolar apparatus which blackens on impregnation with osmic acid, and which seems to possess some degree of permanence from vacuole to vacuole and in some cases from parent to daughter cell. Remarkably little is known about the mechanism of systole. General body turgor may contribute, but it is not essential. In ciliates the main force is local and probably comes from a tension in the wall of the vacuole itself, but it is not known whether or not this is an active contraction of an oriented protein layer. The critical process for the initiation of systole is probably the opening of the pore. It is possible that in ciliates there is a rhythmically operating independent timing mechani,sm by which the vacuolar cycle is controlled, but its existence has not been demonstrated.

In spite of the continuing progress of research in the fields of cellular and molecular biology, which has oriented many embryologists towards molecular biology, no concrete explanation of morphogenesis has yet been found. The present state of knowledge of heart development is characterized by an enormous discrepancy between the qualitative descriptions of what happens on the organ level and the more or less quantitative information on subcellular and molecular events. It is generally not understood how cells form tissues and how tissues generate particular forms of an organ. In an attempt to fill the gaps we systematically studied in the period 1968 to 1973 one of the general but rather neglected morphogenetic mechanisms which integrates cells into tissues and organs-cell death. Only a small part of our research on cell death in the development of chick, rat and human embryo hearts has as yet been published in extenso. Most of it has been communicated in papers delivered at different scientific meetings. We would like to use the opportunity offered by Advances to present a syn thesis and integrative review of our results. In this way the actual period of discovery of the existence of cell death and of its morphogenetic role in the heart development come to an end. This opens up the next phase of our research which consists in studies of how cell death is integrated with other morphogenetic mechanisms.

Nowadays the chromosome theory can be presented in much greater detail and with utter confidence, but its two main features remain the same. However, while the role of the chromosomes in heredity and development has been appreciated for a long time, the manner in which they perform their genetic and epigenetic functions has become amenable to critical investigation only in recent years. There is, therefore, still an unmistakable tendency to think of chromosomes in terms of the discrete threads of cell division and, in keeping with this conception, the chromosome cycle is gen erally considered in relation to the microscopically visible changes in morphology which occur during the mechanically active phases of mitosis and meiosis. Chromosome phenotype, however, changes not only during division but throughout the cell cycle. The changes which occur during interphase are, of course, scarcely revealed in morphological modifications of the restless "resting" nucleus. Consequently they are less obvious and correspondingly less amenable to investigation. This accounts for the concentration on the countable karyotype, with its visible properties of pairing and pycnosity, and the measurable movements of separation and segregation.
