Pharmaceutical Strategies to Prevent, Treat, and Recover: Advances and Challenges in Ischemic Stroke and Hemorrhagic Stroke by Yuwen Li,Gang-Min Hur,Tiejun Zhang Pdf

Stroke is one of the leading causes of death and disability worldwide. Among them, ischemic stroke is attributed to an abrupt blockage of blood flow to brain, while hemorrhagic stroke is caused by bleeding into brain. The principal aim of stroke therapy is to decrease mortality and reduce persistent neurologic injury and long-term disability. It also targets to prevent complications and recurrence after stroke as well as minimize the brain function loss caused by the cerebral damage. Fortunately, many strokes could be prevented through healthy lifestyle changes and controlling stroke risk factors, such as cigarette or tobacco smoking, high blood pressure, diabetes, and heart disease. Carotid endarterectomy and stenting of the cervical and intracranial vessels may help reduce recurrent stroke in some cases. Advanced treatments and rehabilitation are helping many stroke patients return to society and their families.

Therapeutic Modulation of the Complement System: Clinical Indications and Emerging Drug Leads by John D. Lambris,Dimitrios C. Mastellos,Edimara S. Reis Pdf

The complement system is a multi-tasking gatekeeper of innate immunity thatintricately interacts with other key defense systems, such as the endothelial barrier, contact activation and coagulation systems, in maintaining tissue immunosurveillanceand homeostasis. Its rapid and forceful activation in the bloodstream not onlyensures the effective containment of microbial infections through potent cytolyticmechanisms, but also alerts the adaptive immune compartment to ensure the mountingof a proper humoral immune response against foreign antigens. However, there isa lurking ‘dark side’ that can lead complement astray, fueling a self-perpetuatingvicious cycle of inflammation, exuberant immune activation and irreversible tissueinjury that collectively exacerbate both acute and chronic pathologies. Indeed, complement dysregulation or excessive activation have been widely recognized askey pathogenic drivers in a wide spectrum of inflammatory or immune-mediateddiseases. Targeted modulation of the complement system at various points ofthe cascade has revealed promising therapeutic targets for ameliorating diseasescores in a number of conditions ranging from ocular, neurodegenerative andthromboinflammatory disorders, to cancer, periodontal diseases, chronic hemolyticanemias, ischemia-reperfusion organ injury, antibody-mediated transplant rejectionand hemodialysis-triggered inflammation. Elegant pre-clinical studies employing a diversified toolbox of highly specificcomplement inhibitors in rodent or primate models of disease have opened newavenues of therapeutic exploration by providing proof of concept for the therapeuticefficacy of complement modulation. At the same time, the clinical experience gainedduring this last decade with the sole complement-specific drug currently in the clinic, eculizumab, has rekindled the interest of biopharmaceutical companies in developingnew and potent complement therapeutics for complement-driven diseases. In this respect, the complement field is witnessing a new surge of clinical trialsthat are evaluating the safety, PK/PD profile and clinical efficacy of promising drugcandidates in a number of clinical conditions driven by complement imbalance orover-activation.

Ayurvedic Herbal Preparations in Neurological Disorders by Murali Muralidhara,Padmanabhan S. Rajini Pdf

Ayurvedic Herbal Preparations in Neurological Disorders provides a comprehensive and mechanistic understanding of the therapeutic potential of many ayurvedic herbal preparations in disease management. Examining research data for evidence-based ayurvedic approaches, this volume begins with a focused introduction to major ayurvedic plants, discussing various mechanisms underlying their neuromodulatory potential in preclinical and clinical settings. Major subsets of ayurvedic plants are discussed, including Bacopa monnieri, Centella asiatica, Withania somnifera, and others. This volume outlines the importance of integrative approaches along with existing treatments in the intervention/management of Alzheimer’s disease, Parkinson’s disease, depressive mood disorders, epilepsy, schizophrenia, and more. More than a mere compilation of studies, this volume identifies relevant gaps for future research avenues and encourages interdisciplinary collaboration and sharing of knowledge to together identify the most efficacious ayurvedic approaches. Highlighting recent developments (e.g., Ayurgenomics, Ayurahar) and other pertinent research for neuroprotection, this book is crucial for anyone researching or working in the field of neurological disorder treatment and prevention. Mechanism and Pharmacodynamic Material Basis of Neurodegenerative Disease Therapies by Zhi Sun,Erxi Wu,Yi Wang Pdf

Neurodegenerative diseases are debilitating disorders defined as loss of function or progressive destructing structure of the nervous system with high prevalence among the aging population. These diseases mainly include Alzheimer’s disease (AD), Parkinson’s disease (PD), Huntington’s disease (HD), and amyotrophic lateral sclerosis (ALS). They are significantly associated with age and degeneration, becoming a devastating global health issue as a result of the growth of mean life expectancy worldwide. Few or no effective treatments are available for age-related neurodegenerative diseases, causing large socioeconomic and personal costs. The last 15 years have seen a boom in the use and integration of “omic” approaches (such as genomic, transcriptomic, epigenomic and metabolomic techniques) to study neurodegenerative disease in an unprecedented way. While, to date, no therapy based on these approaches is available to cure neurodegenerative diseases. Therefore, it is necessary to search for new, safe, and effective drug candidates and treatment strategies. In this Research Topic, our prime focus would be on the use and integration of “omic” approaches to study biomarkers, pathogenesis, disease progression and new therapeutic target of neurodegenerative diseases, so as to search for new, safe, and effective drug candidates.

Novel Technologies Targeting the Rehabilitation of Neurological Disorders by Jie Jia,Jingchun Guo,Lin Yao,Dingguo Zhang Pdf

With the gradual aging of the population, neurological disorders, including stroke, Alzheimer's disease, Parkinson's disease, etc., are causing increasing distress and may even cause the loss of ability to perform activities of daily living. These disorders are generally progressive. Once onset, they may affect the entire life span with subsequent functional impairments such as motor impairment, speech impairment, swallowing impairment, sensory impairment, and cognitive impairment. Rehabilitation therapy is essential for the dysfunction caused by such disorders. Proper rehabilitation programs can improve or compensate for patients' dysfunction, and thus facilitate the restoration of their ability to daily living, help them return to social life, and reduce family and social stress.
