Translational Research Methods for Diabetes, Obesity and Cardiometabolic Drug Development by Andrew J. Krentz,Lutz Heinemann,Marcus Hompesch Pdf

The world is beset by a pandemic of obesity and type 2 diabetes and the need for new drugs is startlingly clear; recent years have seen a huge increase in research activity to fill this gap. The development of new drugs for diabetes and obesity must be founded upon a sound appreciation of the pathophysiology of these common disorders. The dual defects of insulin resistance and impaired insulin secretion are fundamental to the pathogenesis and progression of obesity-associated type 2 diabetes. There is a need to explain how new drugs can counter insulin resistance and insulin deficiency to a broad range of professionals, from clinical scientists active in early (and later) phase drug development to specialist physicians and increasingly primary care doctors who must tailor drug regimens to the individual patient. Clinical research methods for measuring insulin action and insulin secretion have become well-established in proof-of-mechanism studies; however, selection of the best techniques is by no means straightforward. The purpose of the book is to aid the selection of the most appropriate techniques for assessing insulin action, insulin secretion and body composition in humans (with particular reference to new drugs) in phase 1 and 2 studies and aid the understanding of drug effects and non-drug treatment strategies on key biochemical-hormonal defects of obesity and type 2 diabetes. The book will assume a working knowledge of human physiology relating to glucose metabolism and will be of interest to biomedical scientists, pharmacologists, academics involved in metabolic research and clinicians practicing in these specialties.

Clinical Diabetes Research by Michael Roden Pdf

A practical "how to" guide for multiple methods in metabolism, with a critical and objective discussion of strengths, limitations, and appropriate applications of the described methods. Edited by the winner of the Oskar-Minkowski Prize of the EASD in 2006 Clinical trials in populations at risk of or with overt diabetes mellitus are being performed all around the world to test novel drugs and approaches to managing these diseases. During the last decade, new methods and techniques have been introduced – and are being developed further – that facilitate monitoring of metabolism and even diagnosis of certain defects in human metabolism. While some of these methods are restricted to research facilities, others are already used in clinical practice. The correct beneficial use of these methods requires knowledge of the nature of the techniques and their clinical relevance. Clinical Diabetes Research: Methods and Techniques provides summaries of established state-of-the art-techniques and introduces the new technologies, evaluating their potential in clinical diabetes research. One section compares the efficacy of all metabolic tests that measure insulin action. Another section describes methods that allow quantification of whole body and intracellular metabolic fluxes relevant to diabetes research. The final section focuses on the planning and pitfalls of clinical studies in the field of metabolism. Each chapter throughout the book combines the theoretical basis of the approach with practical recommendations. There are further chapters on the design of clinical trials and ethical considerations. The publication is timely in view of the impressive development of the methods and techniques with their application to understanding the pathophysiology of diabetes, insulin resistance syndromes, obesity, and the metabolic effects of treatment. Clinical Diabetes Research: Methods and Techniques is of great relevance for researchers (from students and post-docs to principal investigators) in the field of human metabolism, clinical endocrinologists and diabetologists who are involved in clinical studies.

Translational Research Methods in Diabetes, Obesity, and Nonalcoholic Fatty Liver Disease by Andrew J. Krentz,Christian Weyer,Marcus Hompesch Pdf

This book aims to aid the selection of the most appropriate methods for use in early phase (1 and 2) clinical studies of new drugs for diabetes, obesity, non-alcoholic fatty liver disease (NAFLD) and related cardiometabolic disorders. Clinical research methods to assess the pharmacokinetics and pharmacodynamics of new diabetes drugs, e.g. the euglycemic clamp technique, have become well-established in proof-of-mechanism studies. However, selection of the most appropriate techniques is by no means straightforward. Moreover, the application of such methods must conform to the regulatory requirements for new drugs. This book discusses the need for new pharmacotherapies for diabetes, obesity and NAFLD and the molecular targets of drugs currently in development. Emerging technologies including functional imaging, circulating biomarkers and omics are considered together with practical and ethical issues pertaining to early phase clinical trials in subjects with cardiometabolic disorders. Translational Research Methods in Diabetes, Obesity, and Non-Alcoholic Fatty Liver Disease is of interest to biomedical scientists, pharmacologists, academics involved in metabolic research and clinicians practicing in these specialties.

This timely and clinically oriented book uniquely addresses the entire spectrum of treatment for severe obesity and diabetes. It thoroughly examines all aspects from robotic surgery to cell therapy, from the latest antidiabetic drugs to endoscopic procedures and devices, from appetite-inhibiting signals and nutrients to gastric arterial embolization. It offers objective and authoritative information for practicing professionals; not only are successful interventions discussed and updated, but cutting-edge and experimental pathways are also anticipated. Obesity is the foremost nutritional challenge worldwide. Its companion is type-2 diabetes, the most prevalent metabolic disease. Together they represent the diabesity epidemic, which is draining resources, overwhelming healthcare facilities, and impacting life expectancy and quality of life everywhere. No country has been spared, and most medical specialties have been affected. Many books focus on the surgical options for obesity and diabetes, namely bariatric and metabolic operations. Information on the clinical and pharmacological treatment is also available, conveyed by standard texts on internal medicine and endocrinology. Yet endoscopic maneuvers and prostheses, cell therapy for diabetes, and other innovative approaches are hard to come by, as conventional texts overlook such therapies. This timely publication bridges the gaps between all different therapeutic options, also covering advances in the conservative, surgical and parasurgical domains. This book will appeal to seasoned specialists, students and healthcare professionals in training alike.
