Cytokines are soluble proteins secreted by a wide variety of cells in the body, playing crucial roles in many physiological responses and are involved in the pathophysiology of cardiovascular diseases (CVDs). Recently, there is great progress in discoveries that some novel cytokines, including secreted frizzled-related proteins (SFRPs), adiponectin, and C1q/TNF-Related Proteins (CTRPs), can perform a protective effect in cardiometabolic diseases, heart failure and coronary heart disease. However, the mechanisms underlying the molecular and pathophysiological actions of these cytokines in CVDs are still vague. Novel mechanisms of cell death including pyroptosis, autophagy, ferroptosis, cuproptosis, and PANoptosis have been considered novel therapeutic targets for CVDs. Some cytokines have been shown to regulate the cell death pathway in the incidence and development of CVDs. Further investigations are needed to understand the link between these novel cytokines and novel cell death models and pathways in CVDs.

In the following paragraphs, the reader will find an overview of the contents of this volume. Greene and Robb review data they have generated over the past 2-3 years with respect to characterization of hormone-specific Interleukin-2 (IL-2) recep tors on the surface of activated T cells. Their chapter traces the development of a quantitative assay for assessment of IL-2 receptors based on the preparation and use of radiolabeled IL-2 prepared biosynthetically with the aid of IL-2-pro ducer leukemic cells. The authors then describe an alternate approach, the prep aration of a monoclonal antibody previously determined to be directed against a T -cell-activation antigen. This so-called anti-Tac antibody was later found to recognize a determinant on or near the IL-2 receptor.

Substances released from activated immune cells (cytokines) stimulate peripheral nerves, thereby signaling the brain and spinal cord that infection/inflammation has occurred. Additionally, peripheral infection/inflammation leads to de novo synthesis and release of cytokines within the brain and spinal cord. Thus, cytokines effect neural activation both peripherally and centrally. Through this communication pathway, cytokines such as interleukin-1, interleukin-6 and tumor necrosis factor markedly alter brain function, physiology and behavior. One important but underrecognized aspect of this communication is the dramatic impact that immune activation has on pain modulation. The purpose of this book is to examine, for the first time, immune-to-brain communication from the viewpoint of its effect on pain processing.
