GHRH, GH, and IGF-I by Marc R. Blackman,S. Mitchell Harman,Jesse Roth,Jay R. Shapiro Pdf

Recent research suggests that adult growth hormone (GH) deficiency, whether of pathological or physiological origin, is associated with a dis tinct syndrome that includes alterations in body composition, endocrine metabolic function, immune competence, and physical and psychosocial well-being. Not surprisingly, substantial investigative effort is currently focused on validating the above hypothesis and on determining whether restoration of a normal GH-IGF-I axis in various states of adult GH deficiency is clinically useful, safe, and cost-beneficial. This book contains the proceedings from the Symposium on GHRH, GH, and IGF-I: Basic and Clinical Advances, held December 9 to 12, 1993, in San Diego, California, and sponsored by Serono Symposia USA, Inc. The conference was meant to highlight selected novel and exciting clinical research developments related to possible therapeutic uses of recombinant human GH and IGF-I, GHRH, GH releasing peptides, and related GH secretagogues. This meeting occurred only one year after a similar Serono symposium that was somewhat more oriented to novel basic science discoveries, thus attesting to the current scientific and clinical interest in this area.

Gigantism and Acromegaly by Constantine A. Stratakis Pdf

Gigantism and Acromegaly brings together pituitary experts, taking readers from bench research, to genetic analysis, clinical analysis, and new therapeutic approaches. This book serves as a reference for growth hormone over-secretion and its diagnosis and treatment for endocrinologists, pediatricians, internists, and neurosurgeons, and for geneticists. Pharmaceutical companies may use it as a reference for drug development and research. Students, residents and fellows in medicine and endocrinology and genetics will also find it valuable as it provides a single up-to-date review of the molecular biology of gigantism and acromegaly as well as recommended approaches to evaluation and management. Acromegaly is a rare pituitary disorder that slowly changes its adult victim’s appearance over time: larger hands and feet, bigger jaw, forehead, nose, and lips. Generally, a benign pituitary tumor is the cause and symptoms of acromegaly can vary from patient to patient, making a diagnosis difficult and prolonging suffering for years. Early detection is key in the management of acromegaly as the pathologic effects of increased growth hormone (GH) production are progressive and can be life-threatening as the result of associated cardiovascular, cerebrovascular, and respiratory disorders and malignancies. Accessible, up-to-date overview of the characteristics, state-of-the-art diagnostic procedures, and management of acromegaly and gigantism Provides a unique compendium of endocrinology, genetics, clinical diagnosis and therapeutics Contains contributions from internationally known experts who have treated patients with acromegaly and gigantism

Growth Hormone Deficiency in Adults by Jens O. L. Jørgensen,Jens Sandahl Christiansen Pdf

It has been known for over 40 years that GH-deficient-children benefit from replacement with the hormone. But GH, essential for longitudinal growth, also plays a role after completion of final height. With the introduction of biosynthetic human GH 20 years ago, the use of GH was no longer restricted to severe growth retardation in hypopituitary children. This book will take the reader behind the myths of GH and into the real world of clinical endocrinology. The contributions stem from recognized clinicians and scientists who have been working in the field for decades. The contents encompass traditional end points of GH therapy such as body composition, bone biology and physical performance. Attention is also devoted to diagnostic aspects and side effects. Additional features range from clinical epidemiology to quality of life, and novel areas such as the impact of traumatic brain injury on pituitary function are also covered. The present volume of Frontiers of Hormone Research is essential reading for health care professionals interested in clinical endocrinology and GH.

Growth Hormone Secretagogues by E. Ghigo Pdf

Growth hormone secretagogues (GHS) administered alone or in combination with growth hormone releasing hexapeptides, are effective probes for the diagnosis of GH deficiency in both children and adults. Current research has developed and tested different GHS compounds that are active by the oral route, and have improved potency and bioavailability, giving rise to exciting therapeutic possibilities. There was an enthusiastic response from experts in this area to the idea of distilling the huge amount of available data into one multi-authored volume. Each contributor has advanced the field of knowledge, and has here emphasized the practical aspects of their work, reviewing the subject in the light of their own experience. Therefore, the theme of the book is a practical one. The volume deals with all aspects of GHS that are relevant to the field, from the chemical structure to the different analogues, to the cloning and expression of the GHS-receptor and the role of these compounds in the physiological control of GH secretion. Also discussed are the most recent advances in relation to the possible role of these compounds in the diagnostic therapeutic settings in different clinical situations, either in children, adults or the elderly. The book meets the requirement of covering most, if not all of the advances in the field. It will enable scientists and clinicians to keep abreast of the rapidly evolving knowledge of the most recent years, and will also prove useful as a review for all interested in this topic.

Human Growth Hormone Pharmacology by Kathleen T. Shiverick,Arlan L. Rosenbloom Pdf

This book addresses a wide range of basic and clinical issues in the physiology and pharmacology of growth hormone. The volume is organized like a textbook. It begins with factors contributing to GH gene expression, the functional relationships of the GH receptor, molecular biological analysis of the GH-GH receptor complex and proceeds to describe the insulin-like growth factor axis. Transgenic models are discussed for analysis of discrete effects. These discussions provide a bridge to clinically oriented discussions of growth abnormalities in GH deficient children, GH insensitivity due to deficiency of the GH receptor, and the concept of aging as a GH/IGF-I deficiency state. Discussions also include the immune system as a source and a responder to GH, GHRH, IGF-I and the effects of GH excess.

Growth Hormone Secretagogues by Barry B. Bercu,Richard F. Walker Pdf

The traditional concept of a neuroendocrine mechanism for regulation of growth hormone (GH) secretion is based in large part on the work of Roger Guillemin. The work of Dr. Guillemin, who was awarded the 1977 Nobel Prize in Physiology and Medicine, supported the view that quantita tive change in GH secretion was the net result of pituitary stimulation and inhibition by the hypothalamic neurohormones, GH releasing hormone (GHRH), and somatostatin (somatotropin release inhibiting factor; SRIF), respectively. During the 1970s, another endocrine research pioneer, Dr. Cyril Bowers, discovered that structural modification of enkephalin re sulted in a family of peptides with GH releasing properties. These com pounds, simply called GH releasing peptide (GHRP), were originally thought to mimic GHRH. However, upon subsequent investigation they were found to supplement the activity of the natural hormone through a different mechanism. Nearly two decades after their discovery, the differ ences between GHRP and GHRH have been described by many different laboratories throughout the world. The complementary GH secretagogues have different binding sites, second messengers, and effects on gene expres sion. Based on these differences, it has been suggested that expansion of the original two hormone mechanisms for GH regulation to include a third molecule may be appropriate, even though the naturally occurring ana logue of GHRP has not yet been identified. Despite our lack of knowledge concerning the natural product mimicked by GHRP, clinical development of the new family of GH secretagogues for diagnostic and therapeutic purposes has begun in earnest.

Growth Hormone II by Barry B. Bercu,Richard F. Walker Pdf

This book is a compendium of proceedings from the Symposium on Growth Hormone II: Basic and Clinical Aspects. The intent of the sym posium was to review current aspects of the rapidly expanding science of growth hormone (GH) neuroendocrinology, especially those develop ments that occurred since our last meeting in 1985. The meeting was timely since there have been many new and exciting developments in basic research on the GH axis, and several new clinical applications for GH other than growth have been identified. For example, significant strides have been made into use of the bioengineered hormone as an anabolic in debilitated states and as an immunorestorative agent. Both these areas have received most attention as they relate to geriatric popu lations. Furthermore, recent focus on a novel family of GH secretagogues that complement the action of GHRH has shown that in animal models, these compounds have the potential to restore GH secretion in deficient or insufficient states, including growth retardation, obesity, and aging. Accordingly, these xenobiotics may have significant potential as drug products to stimulate physiological GH secretion and, thus, resist or reverse some of the maladaptive changes in form and function resulting from GH insufficiency. The importance of these agents as drug candidates is underscored by the diversity of molecules that have been studied by various pharmaceutical companies and the recent discovery that non peptide analogs with high oral bioavailability can be synthesized.

New Human Growth Hormone Research by George V. Artwelle,Francis G. Wislon Pdf

Growth hormone (GH) or somatotropin (STH) is a protein hormone that stimulates growth and cell reproduction in humans and other animals. It is a 191-amino acid; single chain polypeptide hormone which is synthesised, stored, and secreted by the somatotroph cells within the lateral wings of the anterior pituitary gland. HGH promotes growth in children and plays an important role in adult metabolism. The body secretes the hormone, in decreasing amounts, throughout our lifetimes. The amount of hormone in the body can be measured by levels of IGF-1 (Insulin Growth Factor). Growth hormone has a profound effect on all the cells of the body, more than any other hormone because it is the cell generator. This book presents the latest research in the field.
