In a sense, propionic acid bacteria are domesticated bacteria. They might have been used for cheese making as early as 9000 years Be. In the last 40 years their pmctical uses have expanded to include vitamin B12 and propionic acid production, bread baking, starters for ensilage and some pharmaceutical prepamtions. New prospects for their future uses are also emerging, based on the useful properties recently discovered. This monograph is the result of many years of investigating propioni bacteria by the faculty, staff and postgraduate students in the Department of Microbiology at the Moscow State University, as well as a number of scientists in other countries. The encouragement and various contributions of my colleagues has made this book possible, which might as well be entitled "My life with propionic acid bacteria", since these bacteria were the subject of our investigations for more than 40 years. Corynebacteria are a diverse group Gram-positive bacteria found in a range of different ecological niches such as soil, vegetables, sewage, skin, and cheese smear. Some, such as Corynebacterium diphtheriae, are important pathogens while others, such as Corynebacterium glutamicum, are of immense industrial importance. In fact C. glutamicum is one of the biotechnologically most important bacterial species in use today with an annual production of more than two million tons of amino acids, mainly L-glutamate and L-lysine. Due to its industrial importance, C. glutamicum has been studied extensively over the years, and the publication of the C. glutamicum genome sequence in 2003 provided renewed impetus to these studies. To date, the complete genome sequences of four different species have been published, and sequencing of at least two more species is ongoing. These genomic data have enabled a dramatic improvement in our understanding of the corynebacterial genome architecture, metabolic processes, species-specific traits, industrial capabilities, and potential roles in pathogenicity of humans and animals. In addition these genome sequences, allied with newly developed genetic tools will consolidate C. glutamicum as a model organism for systems biology. Research in this area has never been more exciting. This volume brings together the expertise and enthusiasm of today's leading experts to provide a description of the current state-of-the-art molecular biology toolbox available for Corynebacterium research, including global analyses techniques such as comparative genomics, transcriptome, proteome and metabolome analysis as well as the most recent knowledge on Corynebacteriumpromoter structures and vector systems. In his introduction to the Ciba Foundation Symposium on "Immunopotentia tion," Sir Peter Medawar stated that, "for the past twenty years the control of the immune response has been virtually equated to immunosuppression because the great goal of applied immunology has been the transplantation of tissues," and that, "with the discovery of tumor immunity-the focal point of immuno logical control has changed from immunosuppression to immunpotentiation and correspondingly the great prize of applied immunology has become the preven tion and control of malignant growth." Corynebacterium parvum is now univer sally recognized as being the most potent presently known immunopotentiator. C parvum is at the center of almost all papers presented at this conference and it has been analyzed from various viewpoints: taxonomy, pharmacology, immunopotentiation, stimulation of host resistance to tumor invasion in experi mental models and in human therapeutics. Almost all data reported in this volume are original and recently obtained. The discussions were spontaneous, stimulating, and highly enriching, and the results already established and re ported here are in many respects unique. Nonspecific immunopotentiation has strongly penetrated into immunology, immunopathology, and, recently, oncology.
