Late-onset Alzheimer disease (LOAD) is the most common cognitive disorder affecting over five million Americans. Over the past two decades, hundreds of variations in over one hundred twenty candidate genes have been reported to be associated with LOAD. However, only the apolipoproteinE-4 allele has been widely accepted as a risk factor. The deluge of reported susceptibility alleles, lack of systematic follow-up, limited resources, small sample sizes, and varying experimental designs have complicated the search for confirmed LOAD genetic variants. Whole genome approaches are gaining popularity, however large mega-base gaps remain within the available assays so comprehensive coverage in some critical regions of the genome is not possible. In this study, we utilize the Genomic Convergence approach to narrow our interrogation to variations within nineteen candidate genes with the best evidence for involvement in LOAD risk. Candidate genes were selected based on published identification of each gene by at least three different experimental methods (linkage analysis, candidate gene association and gene expression). Using dense SNP genotyping, we examine candidate variants for association with LOAD in a large case-control LOAD dataset that is independent from those used to generate each of the antecedent association reports.

Paternal age and epigenetic changes appear to have an effect on schizophrenia and epigenetic silencing has been associated with several of the progeroid syndromes of premature aging.
