Modern Analytical Ultracentrifugation by Todd M. Schuster,Thomas M. Laue Pdf

There are numerous examples in the history of science when the parallel develop ments of two or more disciplines, methodologies, technologies or theoretical in sights have converged to produce significant scientific advances. The decades following the 1950s have produced several such significant advances, as a result of a convergence of developments in molecular biology and in solid state-based electronics instrumentation. Since one of these areas of significant advancement, analytical ultracentrifu gation, has been undergoing a renaissance, we thought it would be a useful activity to call upon a group of researchers who have been developing either the experi mental or theoretical aspects of the methodology and gather in one place a group of articles summarizing the current status of the field. The success of recombinant DNA methodologies at producing biologically active macromolecules of commer cial interest has evoked interests in mechanisms of function. Pursuit of the related questions has emphasized the importance of studies of macromolecular binding and interaction. Several contributions to this volume remind us that analytical ultra centrifugation is rigorously based on solid thermodynamic theory and, as such, is fully capable of providing comprehensive quantitative descriptions of molecular interactions in solution. Furthermore, a number of the chapters provide examples, along with innovative methods for carrying out these characterizations. The past decade has seen several developments that reflect the rebirth of interest in analytical ultracentrifugation.

Sedimentation Velocity Analytical Ultracentrifugation by Peter Schuck,Huaying Zhao Pdf

Analytical ultracentrifugation is one of the most powerful solution techniques for the study of macromolecular interactions, to define the number and stoichiometry of complexes formed, and to measure affinities ranging from very strong to very weak and repulsive. Building on the data analysis tools described in the volume Sedimentation Velocity Analytical Ultracentrifugation: Discrete Species and Size-Distributions of Macromolecules and Particles, and the experimental and instrumental aspects in the first volume Basic Principles of Analytical Ultracentrifugation, the present volume Sedimentation Velocity Analytical Ultracentrifugation: Interacting Systems is devoted to the theory and practical data analysis of dynamically coupled sedimentation processes. This volume is designed to fill a gap in biophysical methodology to provide a framework that builds on the fundamentals of the highly developed traditional methods of analytical ultracentrifugation, updated with current methodology and from a viewpoint of modern applications. It will be an invaluable resource for researchers and graduate students interested in the application of analytical ultracentrifugation in the study of interacting systems, such as biological macromolecules, multi-protein complexes, polymers, or nanoparticles.

CRC Handbook of Viruses by Horace M. Mazzone Pdf

As a distinct class of macromolecules, viruses are continually being studied in order to determine their properties. Following a knowledge of host-range infectivity, the particle mass-molecular weight of the virus and related properties - including size, shape, sedimentation, and diffusion coefficients - are also important characterizations. In the literature, these values have been determined for many viruses, and a variety of techniques are available by which such properties may be analyzed. Until now, there has been no single source for such information that the interested investigator may consult, and no databases provided this kind of information. CRC Handbook of Viruses: Mass-Molecular Weight Values and Related Properties corrects this deficiency by presenting such data for all classes of viruses; centering on viruses, their molecular weight, and their related properties; and acquainting the investigator to many methods for obtaining the mass-molecular weight value of viruses. This singular study explains the variety of methods available to the researcher as well as provides examples of each method. Molecular weight values are accessed directly from the book, saving the investigator a tedious search through the range of literature. Chapters discuss: Viruses as infectious agents and their role in establishing the relatively new discipline of molecular biology Essential components of viruses, protein, and nucleic acid - considering their discovery, nature, structural organization, and the forming of viruses from nucleoproteins Basic aspects of virus purification, preparative centrifuge, and various purification methods Basic aspects of crystallography, including procedures, x-ray analysis of the viral protein component, the arranging of subunits, and the composition of the intact virus Sedimentation experiments and studies used to obtain molecular weight Sedimentation and diffusion coefficients integral to the basic Svedberg equation Sedimentation equilibrium procedures The new Beckman Optima series of analytical ultracentrifuges Scattering studies, including small angle x-ray, small angle neutron, classical light scattering, and electron microscopy The renaissance of instrumentation in classical light scattering techniques Cold neutron facilities currently being set into operation by government laboratories Sizing and solvation of viruses in solution - their "natural environment" Critical modeling experiments Facilities and instrumentation for molecular weight studies - including the high voltage transmission electron microscope for obtaining mass values of viral inclusion bodies CRC Handbook of Viruses serves the: Researcher seeking values of virus molecular weight and related parameters Investigator getting started in virology and seeking information on physical chemical procedures Student interested in viruses as infectious agents

A follow-up to the experimental and instrumental aspects described in Basic Principles of Analytical Ultracentrifugation, the volume Sedimentation Velocity Analytical Ultracentrifugation: Discrete Species and Size-Distributions of Macromolecules and Particles describes the theory and practice of data analysis. Mathematical models for the sedimentation process and the evolution of detected signals are developed in a comprehensive framework, jointly with the description of current and historical strategies for how to extract from noisy experimental data the physical parameters of interest, such as size, mass, and shape, composition, and polydispersity of sedimenting particles. The methods are extensively illustrated, and supported with practical applications, as well as cross-references where to find the methods in the public domain software SEDFIT and SEDPHAT. The systems covered are discrete or polydisperse mixtures of sedimenting molecules or particles in dilute solution, such as proteins and other biomolecules and their stable complexes, man-made polymers, and nanoparticles, observed in different optical systems. A useful reference for researchers and graduate students of macromolecular disciplines, these methods form the essential foundation for the analysis of dynamic interacting systems, which are covered in the volume Sedimentation Velocity Analytical Ultracentrifugation: Interacting Systems. Basic Principles of Analytical Ultracentrifugation by Peter Schuck,Huaying Zhao,Chad A. Brautigam,Rodolfo Ghirlando Pdf

Analytical ultracentrifugation (AUC) can supply rich information on the mass, shape, size distribution, solvation, and composition of macromolecules and nanoscopic particles. It also provides a detailed view of their reversible single- or multi-component interactions over a wide range of affinities. Yet this powerful technique has been hard to master in mainstream molecular sciences due to a lack of comprehensive books on the subject. Filling this gap in the literature of biophysical methodology, Basic Principles of Analytical Ultracentrifugation explains the fundamentals in the theory and practice of AUC. The book provides you with up-to-date experimental information to confidently practice AUC. You will understand the basic concepts, full potential, and possible pitfalls of AUC as well as appreciate the current relevance of past work in the field. The book first introduces the basic principles and technical setup of an AUC experiment and briefly describes the optical systems used for detection. It then explores the ultracentrifugation experiment from a macromolecular standpoint, offering a detailed physical picture of the sedimentation process and relevant macromolecular parameters. The authors present important practical aspects for conducting an experiment, including sample preparation, data acquisition and data structure, and the execution of the centrifugal experiment. They also cover instrument calibration and quality control experiments.
