Quinolone Antibacterials by Jochen Kuhlmann,A. Dalhoff,H.-J. Zeiler Pdf

It has been over 30 years since the first clinically important member of the quinolone class, nalidixic acid, was introduced into medical practice. The modification produced in the quinolone nucleus by introducing a fluorine at the 6-position led to the discovery of the newer fluoroquinolones with enhanced antibacterial activities as compared to nalidixic acid. By now a great deal of preclinical and clinical experience has been obtained with these agents. The intense interest in this class of antibacterial agents by chemists, micro biologists, toxicologists, pharmacologists, clinical pharmacologists, and clini cians in various disciplines encouraged us to summarize the information on the history, chemistry, mode of action and in vitro properties, kinetics and efficacy in animals, mechanisms of resistance, toxicity, clinical pharmacology, clinical experience, and future prospects in one volume of the Handbook of Experimental Pharmacology. As this series deals predominantly with "experimental" characteristics of drugs, our volume is dedicated specifically to quinolones and emphasizes principally their preclinical and clinical phar macological characteristics, despite the existence of several summaries on quinolones. The chemistry of the quinolones is described in detail. The chapter on the mode of action of quinolones reports the conclusive evidence that gyrase is the intracellular target of the quinolones; however, another enzyme, topoisomerase IV, may also be a target for quinolones, and the exact mechanisms by which quinolones act bactericidally are far from being understood.

Quinolone Antimicrobial Agents by David C. Hooper,John S. Wolfson Pdf

Quinolones remain the most important class of antimicrobial agents discovered in recent years – over 1000 have been synthesized and evaluated. Since the publication of the original edition, considerable strides have been made in the research on structure–activity relationships, mechanism of action, resistance, pharmacodynamics and drug interactions. This edition consolidates and substantially updates our current state of knowledge of quinolones, with thirteen new chapters having been added.

The 4-Quinolones: Anti Bacterial Agents in Vitro by Geoff C. Crumplin Pdf

The 4-quinolone antibacterial agents have been under development and in use for over 25 years, but despite an enormous body of research work there are still many basic questions that remain unanswered. For example, we do not know how these agents kill susceptible cells - or even how the drugs interact with the target mol- ecule(s). However, we do know that the 4-quinolones display many properties which are not shared with any other group of antibacterial agents. This book raises the unanswered questions and assesses the potential significance to future chemotherapy of their unique features. The book also raises matters which are significant for physicians as well as for the pharmaceutical industry and the assessment of these agents by legislative bodies concerned with drug safety.

The quinolones are making a significant contribution to patient care and have added a new dimension to antibacterial therapy. During the last decade, they became important agents for treating hospital-acquired infections. This volume covers their mechanisms of action, resistance mechanisms and epidemiology, pharmacokinetics, safety and clinical applications. Their effectiveness for respiratory, urinary, sexually transmitted, skin, soft tissue, bone and joint infections, infections of the eyes, ears, nose and throat, and less common infections such as mycobacterial, Brucella and others are reviewed. Also described is their use in pediatric, oncology and immunocompromised patients. The quinolone story continues to be intriguing with exciting future implications. This monograph is an important comprehensive review and update for researchers and clinicians in pharmacology, clinical medicine, molecular and cell biology, epidemiology and the pharmaceutical industry.

New Trends in Allergy III by Johannes Ring,Bernhard Przybilla Pdf

The international symposium "New Trends in Allergy", held in Munich from July 13 to 15, 1990, brought together for the third time since 1980 some of the most experienced researchers working in the field of allergy. This volume comprises the papers presented at this meeting. All over the world, and not merely in the industrialized countries, allergy is becoming a cause of evermore serious diseases. In recent years, research in the field of allergy has provided numerous impor tant and fascinating results extending our knowledge considerably. Despite the new insights into basic mechanisms of allergic reactions, improved diagnostic methods, and new therapeutic approaches, how ever, many questions remain to be answered, including: Are allergies really increasing in frequency? If so, what are the reasons? Especially, does environmental pollution playa role? Which factors influence IgE synthesis? Can the IgE immune response be switched off? Does the nervous system interact with allergic reactions? If so, what are the mechanisms? Are new approaches in allergy prophylaxis and allergy therapy effi cient? What measures have proven useful and deserve to be employed in daily practice? In this volume, these questions and other current topics are dealt with. As each issue is covered by authors competent in the respective fields, the result is an extensive and critical review of the state of the art. Going through these papers, one comes to the conviction that allergy research is a multifacetted, explosively expanding, most stimulating field of work.

Antibacterial Chemotherapeutic Agents by S.L. Dax Pdf

Over the past 50 years a wide variety of antibacterial substances have been discovered and synthesised, and their use in treating bacterial infection has been spectacularly successful. Today there are several general classes of antibacterial drugs, each having a well established set of uses, and together they form the mainstay of modern antibacterial chemotherapy. In search for new and improved agents, the pharmaceutical researcher needs to be well informed on many topics, including existing agents, their modes of action and pharmacology, and possible synthetic approaches. In this new book the author has brought together a wide range of information on the principal classes of antibacterial agents, and he covers, for each group, their history, mode of action, key structural features, synthesis and bacterial resistance. The result is a compact and concise overview of these very important classes of antibacterial agents.

Antimicrobial Chemotherapy by Roger Finch,Peter Davey,Mark H. Wilcox,William Irving Pdf

Antimicrobial agents are essential for the treatment of life-threatening infections and for managing the burden of minor infections in the community. In addition, they play a key role in organ and bone marrow transplantation, cancer chemotherapy, artificial joint and heart valve surgery. Unlike other classes of medicines, they are vulnerable to resistance from mutations in target microorganisms, and their adverse effects may extend to other patients (increased risk of cross-infection). As a consequence, there is a constant requirement for new agents, as well as practices that ensure the continued effective prescribing of licensed agents. Public awareness and concerns about drug resistant organisms has led to widespread publicity and political action in the UK, Europe and worldwide. The control of drug resistance and the implementation of good prescribing practice are now legal requirements in the UK as a result of the UK Health Act (2008). These fundamental changes underscore the need for a thorough understanding of the advantages and risks associated with specific antibiotic choices. This sixth edition of Antimicrobial Chemotherapy continues to be a valuable resource for undergraduates and graduates requiring a thorough grounding in the scientific basis and clinical application of these drugs. This new edition is updated to include the most recently licensed agents, notably in the treatment of viral infections including HIV/AIDS, and contains new guidance on prescribing practice and infection control practices that limit the development and spread of resistant organisms.
