since the earliest results on the linkage between ras activation and cell transformation appeared a vast amount of additional information has been generated which emphasizes the importance of ras-related genes in membrane trafficking, cell proliferation, differentiation and cancer. These advances led to the development of new strategies for the diagnosis, prognosis and prevention of cancer. Ras-related genes appear to be central to the mechanism of transformation by other oncogenes and therefore constitute a focal point upon which attempts to intervene in the process of carcinogenesis will be concentrated. The present volume contains the contributions to the NATO Advanced Research Workshop on the "The Superfamily of ras-Related Genes" held in Agia Pelagia, Heraklion, Crete, Greece, May 1991. At this meeting the leading researchers in this field carne together to discuss the most recent progress in analysis of biological function of the ras-related genes. As an organizer I feel that the Workshop was a success, and would like to thank all the participants and the contributors to this volume. "Cancer viruses" have played a paradoxical role in the history of cancer research. Discovered in 1911 by Peyton Rous (1) at the Rockefeller Institute, they were largely ignored for several decades. Witness his eventual recognition for a Nobel Prize, but not until 1966-setting an all time record for latency, and testimony to one more advantage of longevity. In the 1950s, another Rockefeller Nobelist, Wendell Stanley, spearheaded a campaign to focus attention on viruses as etiological agents in cancer, his plat form having been the chemical characterization of the tobacco mosaic virus as a pure protein-correction, ribonucleoprotein-in 1935 (2). This doctrine was a centerpiece of the U.S. National Cancer Crusade of 1971: if human cancers were caused by viruses, the central task was to isolate them and prepare vaccines for immunization. At that point, many observers felt that perhaps too much attention was being devoted to cancer viruses. It was problematic whether viruses played an etiological role in more than a handful of human cancers.
The GTPase switch appears to be almost as old as life itself, and nature has adapted it to a variety of purposes. This two-volume work surveys the major classes of GTPases, including their role in ensuring accuracy during protein translation, a new look at the trimeric G-protein cycle, the molecular function of ARF in vesicle coating, the emerging role of the dynamin family in vesicle transfer, GTPases which activate GTPases during nascent protein translocation, and the many roles of ras-related proteins in growth, cytoskeletal polymerization, and vesicle transfer. 80 chapters contain much previously unpublished data and, at the rate the extended family of GTPases is growing, it is unlikely that it will again sit for a group portrait such as this. The first edition of Oncogenes (1989) focused on several of the better known transforming mechanisms and surveyed a spectrum of solid tumors and hematologic malignancies. Several of the nearly 50 known oncogenes most relevant to human disease were examined. In contrast, this volume presents a very different profile and balance of subject material that reflects the rapidly changing field of molecular oncology and its newly emerging concepts. Among the most important discoveries of the past 4 years are the identification of nearly a dozen different tumor suppressor genes and the finding of an entirely new class of cancer-causing gene (bcl-2) that acts by inhibiting cell death rather than stimulating cell proliferation. This edition begins by reviewing selected malignancies in which our earlier search for clinically relevant oncogenes has led to more focused studies on gain-of-function and loss-of-function genetic abnormalities, as well as autocrine and paracrine growth factor loops known to regulate tumor physiology and malignant cell behavior. Curiously, many of these genetic and functional abnormalities are shared by several different tumor types and are not uniformly present in all tumors of the same type. This observation brings up molecular questions about the tissue-specific determinants that underlie individual cancers and also gives added impetus to the suggestion that molecular abnormalities (referred to as tumor markers) be included among the histopathologic features used for clinical diagnosis and manage ment.
Discoveries that a small subset of cellular genes --- proto-oncogenes --- are frequent targets of carinogenic insults or chance that convert them to oncogenes have opened new areas of approach to understanding and eventually controlling the events leading to cancer. New technology has made it possible not only to identify genes involved in the etiology of cancer, but to dissect their molecular composition. This symposium brought together scientists from areas of carcinogenesis and molecular biology to review the state-of-the-art in this field. The papers presented in this proceedings volume can be grouped into four broad areas: the mechanisms of onc gene activation and associations of onc genes with tumours, the mechanisms of onc gene action, the physiology of proto-onc genes, and the discovery and characterization of new onc genes.
