The cytochemical bioassay system was described in a short abstract in 1971, and more fully, in the cytochemical bioassay of corticotrophin, in 1972. Since then, cytochemical bioassays have been described for several polypeptide hormones, and these assays are already widely used. It is expedient that the subject should be reviewed, as it is in this monograph, by one writer who has had the good fortune to have taken part in the growth of cytochemistry from its early origins to its present position as the basis of possibly the most sensitive bioassay system currently available. However, it should be noted that major contributions have been made by many, both to the development of the subject and to the establishment of the bioassays. The object of this preface is to try to give some perspective to the growth of this subject and to record that the cytochemical bioassay system has been fostered by many outstanding scientists in an atmosphere of remarkable goodwill. Histochemistry and cytochemistry are essential tools in biomedical research and routine service laboratories. Most texts on histochemistry fall into one of two categories: 1. Encyclopaedic texts covering all or nearly all information available on the whole or selected parts of histochemistry. 2. Reviews or surveys of methods found to be useful by the author(s). While the former category often appeals to the more philosophically inclined reader, direct guidance on the selection of technique may be difficult to find. In contrast, the latter category are often excellent sources for details on how to perform a particular method with a reasonable chance of success. Consideration of the exact mechanism of staining, of possible reasons for failure, and of alternative techniques are, however, frequently lacking. This book is an introduction to the scientific basis of histochemistry and is intended to provide a background for the selection and development of appro priate methods. It is not a "cook book" and readers expecting exhaustive methodological descriptions will be disappointed. Although most ofthe contributors to this book would not describe themselves as histochemists, they have all at some time found it essential to develop a basic understanding of histochemistry. o history of endocrinology can be written without reference N to Sir Humphry Davy Rolleston, whose monumental study of the subject appeared in 1936 under the modest subtitle: The Endocrine Organs in Health and Disease with an Historical Review. It was based on the author's Fitzpatrick Lectures at the Royal College of Physicians of London in 1933 and 1934. The lectureship, which dates from 1901, is devoted to the History of Medicine. Rolleston's work as regards scholarship and delivery cannot be surpassed and will remain the solid basis for any further study. It is of interest to note that Rolleston gave the Fitzpatrick Lectures when he was 71 years of age and had his book published when he was 74. By that time he had achieved most of his professional aims and all the honours a distinguished medical career can offer (see Section II). He perceived clearly that endocrinology was "an enormous subject in a most active stage of growth", which "recently has received most valuable help from organic chemists, who have devoted much time to the elucidation of the structure, isolation and synthesis of the hormones". He remarked that the knowledge of endocrinology was expanding with extreme rapidity, and it has been suggested that in this respect it would appear to be itself influenced by a growth hormone. The incidence of lung cancer has reached epidemic proportions throughout the civilized world. One indication of the dimensions of this problem is that in the United States lung cancer has become the leading cause of cancer death in women as well as men. In 1912 there was a "nearly complete consensus of opinion that primary malignant neoplasms of the lung (were one) of the rarest forms of disease," according to Adler. By 1937, however, it had become clear that the incidence of lung cancer was increasing significantly; this increase has been progressive ever since. It is now well known that some lung cancers give rise to a variety of hormones which, at times, produce clinical manifestations. The association of hormone production with a "nonendocrine" tumor raises many questions, the answers to which may shed some light on the etiology of this prevalent form of cancer. This fascinating prob lem has stimulated a wide variety of studies in both the clinical and the basic sciences. A number of the more recent studies in this field were discussed at the International Symposium on Peptide Hor mones and Lung Cancer held in Marburg, West Germany, on June 18-20, 1984.
