Hepatocellular carcinoma: from personalized medicine to practical guidelines by Sarah El-Nakeep,Anup Kasi,Ahmed Sawabi Shwaky,Wangi Rachel,Htar Aung Pdf

Hepatocellular carcinoma (HCC) is the most common primary liver cancer. HCC is the fourth cause of cancer-related mortality and the fifth most common cancer in the world. The prognosis and survival of patients depend mainly on the cancer stage at time of diagnosis. Earlier detection of the disease has seen improvement in treatment and disease-free survival over the last several decades. Currently, the only curable treatment for HCC is tumor resection or transplantation. These measures are strictly successful only if caught during early-stage HCC. Research has identified multiple molecular pathways involved in HCC on genetic, epigenetic, and proteomic levels. There has been a lot of research into designing drugs to specifically target these molecular pathways affected in patients, and thus contributing to personalized medical treatment. There is abundant bioinformatic research to document the extensive molecular pathways involved as well as drug docking techniques used to assess drug candidates for treating those affected pathways. These bioinformatic techniques result in identifying candidates suitable for further analysis in tissue and animal models and human trials.

An Information Technology Framework for Predictive, Preventive and Personalised Medicine by Leonard Berliner,Heinz U. Lemke Pdf

This book explores how PPPM, clinical practice, and basic research could be best served by information technology (IT). A use-case was developed for hepatocellular carcinoma (HCC). The subject was approached with four interrelated tasks: (1) review of clinical practices relating to HCC; (2) propose an IT system relating to HCC, including clinical decision support and research needs; (3) determine how a clinical liver cancer center can contribute; and, (4) examine the enhancements and impact that the first three tasks will have on the management of HCC. An IT System for Personalized Medicine (ITS-PM) for HCC will provide the means to identify and determine the relative value of the wide number of variables, including clinical assessment of the patient -- functional status, liver function, degree of cirrhosis, and comorbidities; tumor biology, at a molecular, genetic and anatomic level; tumor burden and individual patient response; medical and operative treatments and their outcomes.

Immunotherapy of Hepatocellular Carcinoma by Tim F. Greten Pdf

In this book we provide insights into liver – cancer and immunology. Experts in the field provide an overview over fundamental immunological questions in liver cancer and tumorimmunology, which form the base for immune based approaches in HCC, which gain increasing interest in the community due to first promising results obtained in early clinical trials. Hepatocellular carcinoma (HCC) is the third most common cause of cancer related death in the United States. Treatment options are limited. Viral hepatitis is one of the major risk factors for HCC, which represents a typical “inflammation-induced” cancer. Immune-based treatment approaches have revolutionized oncology in recent years. Various treatment strategies have received FDA approval including dendritic cell vaccination, for prostate cancer as well as immune checkpoint inhibition targeting the CTLA4 or the PD1/PDL1 axis in melanoma, lung, and kidney cancer. Additionally, cell based therapies (adoptive T cell therapy, CAR T cells and TCR transduced T cells) have demonstrated significant efficacy in patients with B cell malignancies and melanoma. Immune checkpoint inhibitors in particular have generated enormous excitement across the entire field of oncology, providing a significant benefit to a minority of patients.

Personalized Medicine in Oncology by Ari VanderWalde Pdf

Nowhere is the explosion in comprehensive genomic testing more evident than in oncology. Multiple consensus guidelines now recommend molecular testing as the standard of care for most metastatic tumors. To aid in the advancement of this rapidly changing field, we intend this Special Issue of JPM to focus on technical developments in the genomic profiling of cancer, detail promising somatic alterations that either are, or have a high likelihood of being, relevant in the near future, and to address issues related to the pricing and value of these tests. The last few years have seen the cost of molecular testing decrease by orders of magnitude. In 2018, we saw the first "site-agnostic" drug approvals in cancer (for microsatellite unstable cancer (PD-1 inhibitors) and NTRK-fusions (TRK inhibitors)). Research on targetable mutations, determination of genetic "signatures" that can use multiple individual genes/pathways, development of targeted therapy, and insight into the value of new technology remains at the cutting edge of research in this field. We are soliciting papers that present new technologies to assess predictive biomarkers in cancer, original research (pre-clinical or clinical) that demonstrates promise for particular targeted therapies in cancer, and articles that explore the clinical and financial impacts of this paradigmatic shift in cancer diagnostics and treatment.

Nanotherapeutics for the Treatment of Hepatocellular Carcinoma by Biswajit Mukherjee Pdf

Hepatocellular carcinoma (HCC) is a leading cause of death globally. Conventional chemotherapeutic agents are unable to penetrate cancerous hepatocytes completely and are toxic to non cancerous cells and tissues. This toxicity significantly compromises the therapeutic outcome of conventional chemotherapeutic agents which is also reflected in the high mortality of the disease. Nanotherapeutics have shed new light onto HCC treatment by enabling site-specific in vivo delivery of chemotherapeutics specifically to neoplastic hepatocytes without affecting normal hepatocytes. Thus, nanotherapeutics have shown considerable potential and there is tremendous impetus for rapid translation from the pre-clinical to the clinical domain to significantly prolong the survival in HCC. In Nanotherapeutics for the Treatment of Hepatocellular Carcinoma, authoritative experts of the field have explored the important aspects of nanotherapeutics against HCC. The book exhaustively, vividly and explicitly describes the molecular pathogenesis, diagnostic aspects and nanotherapy of HCC, while also highlighting the challenges of conventional therapy and the benefits of nanotherapeutics. Chapters of the book also cover recent investigations of nanotherapeutics against HCC, types of nanomedicines, recent patents, commercially available nanotherapeutics and a future perspective to give a comprehensive review of the topic to readers. In addition to these defining features, the book provides several references for further reading. The book is an ideal resource on HCC nanotherapeutics for medical and pharmacology postgraduate students, faculties, researchers, and biomedical scientists working on HCC and nanotherapy.
