The ontogeny of lymphoid cells seems the most appropriate place to start. The early events in T and B cell ontogeny are still confusing. There seems to be no agreement on the data and on the semantics of the question of progenitor vs. stem cells. Nevertheless, we are beginning to understand more about progenitor cells which are committed to particular cell lines, and about stem cells in the sense of having almost unlimited capability of giving rise to undifferentiated progeny. An important future development will be to determine the nature of the substances that attract stem cells and which are produced by specialized thymus epithelium, and perhaps by the bursa. The way stem cells recognize these signals is an important question to answer. Not predictable from mammalian models has been the observation that there is a lack of cells called into the bursa even before the signal for entry of stem cells has been shut off. A likely model suggested that after a certain point in development there were no longer any cells capable of migrating into the bursa and becoming B cells. A fascinating possibility is the suggestion that a cell comes into the bursa, is not committed, then can still wander into the thymus. This cell does not appear to have B cell characteris tics; that is, immunoglobulin is not expressed on its surface.

Polyclonal antibodies, widely used in research and diagnostics, are conventionally isolated from the blood of immunised mammals. The fact that antibodies can also be detected in the yolk of eggs laid by immunised hens, led to the development of the yolk antibody technology as an alternative method less stressful to animals. Hens can be kept under nearly natural conditions and antibodies be isolated from the eggs. Further, the IgY-technology has the advantage that avian antibodies can be produced against several antigens which do not induce a immune response in mammals. Protocols on how to keep and immunise hens and on the extraction, isolation and use of antibodies from yolk are described in detail in this manual. For their work on yolk antibodies the authors received the FISEA (International Foundation for the Substitution of Animal Experimentation, Luxembourg) prize in 1997.

Recent advances in the understanding of the major events that shape the immune recog nition system have been remarkable. The analysis of immunoglobulin (Ig) gene organization and Ig repertoire diversification in lower vertebrates has provided new insight into this process in mammals. Similarly, the understanding of the early development of lymphocytes and of the acquisition of immunological tolerance has been aided by elegant studies in quail/chicken chimeras, using the power of the distinctive markers of the constitutive cells of these birds. Great strides have been made in understanding the role played by major histocompatibility complex (MHC) molecules in antigen presentation and in repertoire selec tion within the thymus. The use of transgenic mice expressing specific T-cell receptor (TCR) genes has elucidated the process of both positive and negative selection. In parallel, there has been considerable progress in our understanding of tolerance, based in part on the use of markers for the V fJ genes of T-cell receptors and in part on the analysis of the behavior of long term T-cell lines. This has led to the realization that both clonal deletion and clonal anergy may play critical roles in the maintenance of unresponsiveness to self antigen. Molecular analysis of the requirements for expression of membrane immunoglobulin molecules has revealed the existence of a complex that appears to be of critical importance in mediating signalling through Ig receptors. In addition, major insights have been obtained into the regulation of expression of genes of immunologic interest.
