Fragment Based Drug Design by Lawrence C. Kuo Pdf

There are numerous excellent reviews on fragment-based drug discovery (FBDD), but there are to date no hand-holding guides or protocols with which one can embark on this orthogonal approach to complement traditional high throughput screening methodologies. This Methods in Enzymology volume offers the tools, practical approaches, and hit-to-lead examples on how to conduct FBDD screens. The chapters in this volume cover methods that have proven to be successful in generating leads from fragments, including chapters on how to apply computational techniques, nuclear magnetic resonance, surface plasma resonance, thermal shift and binding assays, protein crystallography, and medicinal chemistry in FBDD. Also elaborated by experienced researchers in FBDD are sample preparations of fragments, proteins, and GPCR as well as examples of how to generate leads from hits. Contemporary Enzyme Kinetics and Mechanism by Daniel L. Purich Pdf

Selected Methods in Enzymology: Contemporary Enzyme Kinetics and Mechanism provides an introduction to enzyme kinetics and mechanism at an intermediate level. This book covers a variety of topics, including temperature effects in enzyme kinetics, cryoenzymology, substrate inhibition, enol intermediates enzymology, and heavy-atom isotope effects. Organized into 19 chapters, this book begins with an overview of derivation of rate equations as an integral part of the effective usage of kinetics as a tool. This text then examines the practical aspects of initial rate enzyme assay. Other chapters consider the basic procedures used in making decisions concerning kinetic mechanisms from initial-rate data. This book discusses as well the various aspects of both the theoretical background and the applications. The final chapter deals with the importance of achieving proficiency in formulating quantitative relationships describing enzyme behavior. This book is a valuable resource for students and research workers. Enzymologists and chemists will also find this book useful.

Methods in Non-Aqueous Enzymology by Munishwar N. Gupta Pdf

Extending the range of enzymatic catalysis by using non-aqueous media has now developed into a powerful approach in biochemistry and biotechnology. One peculiar feature which distinguishes it from the conventional enzymology (carried out in aqueous buffers) is that the awareness of different parameters that control and influence the behaviour of enzymes in such environments has emerged rather slowly. Science is about being able to repeat what somebody else has done. Absence of knowledge about such well-defined parameters/fac tors has sometimes made some workers rather cautious and diffident about using this approach in their laboratories. But for this, non-aqueous enzymol ogy would be more widely practised. It is these thoughts that made me feel that the availability of some well-defined protocols for various applications invol ving enzymes in non-aqueous environments would further catalyze the growth of this area. Hence this book, in which each chapter has some protocols in a specific area. The protocols are preceded by brief background material. The early chapters, which are of general importance, concern control of water ac tivity and stabilization via immobilization. Some subsequent chapters provide the protocols for transformations involving lipids and carbohydrates, peptide synthesis, and preparation of chiral compounds. The disproportionate focus on lipases is not a coincidence; this class of enzymes has been used more often than others in non-aqueous enzymology.
