Synthetic DNA Delivery Systems by Dan Luo,W. Mark Saltzman Pdf

DNA delivery into cells is a rapidly developing area in gene therapy and biotechnology. Moreover, it is a powerful research tool to determine gene structure, regulation, and function. Viral methods of DNA delivery are well-characterized and efficient, but little is known about the toxicity and immunogenecity of viral vectors. As a result, non-viral, transfection methods of DNA delivery are of increasing interest. Synthetic DNA Delivery Systems is a comprehensive and current resource on DNA transfection. The use of histidine-rich peptides and polypeptides as DNA delivery systems and self-assembled delivery systems based on cationic lipids and polymers are discussed. Targeted delivery to organelles, tumor cells and dendritic cells comprise an important topic.

Molecular Regulation of Endocytosis by Brian Ceresa Pdf

Molecular Regulation of Endocytosis is a compilation of scientific "short stories" about the entry of external substances into cells. As one can see from the chapters, endocytosis regulates diverse processes such as homeostasis of the cell, signal transduction, entry of pathogens and viruses. In addition to the experimental techniques embedded in each chapter, entire chapters are dedicated to experimental approaches that will be useful to all scientists and their model systems. For those more clinically oriented, the final chapters look to the future and ways of utilizing endocytic pathways for therapeutic purposes.

Gene Transfer and Expression Protocols by Edward J. Murray Pdf

Biology is the study of living things. The classical approach might be described as holistic and descriptive, whereas the modern molecular - proach aims to be investigative, reductionist, and mechanistic . Genes contain all the information for the structure of all living things ; thus, the understanding of how genes are regulated is an important step toward understanding the nature of living things. The study of gene regulation has been made more tractable by the design of simple expe- mental models in which a single gene can be isolated from the milieu of the organism. The new science of molecular biology has introduced techniques that permit the design of such experimental models. In - sence, the genome of the organism is dissected in such a manner that specific genes may now be introduced into an appropriate cell line . Subsequent analysis of the proteins expressed from the genes under study results in the identification of the regulatory DNA sequences .

How Free Cationic Polymer Chains Promote Gene Transfection by Yue Yanan Pdf

In this PhD thesis, Yue Yanan addresses a long-overlooked and critical question in the development of non-viral vectors for gene delivery. The author determines that those uncomplexed and cationic polymer chains free in the solution mixture of polymer and DNA facilitate and promote gene transfection. Furthermore, by using a combination of synthetic chemistry, polymer physics and molecular biology, Yue confirms that it is those cationic polymer chains free in the solution mixture, rather than those bound to DNA chains, that play a decisive role in intracellular trafficking. Instead of the previously proposed and widely accepted “proton sponge” model, the author's group propose a new hypothesis based on the results of several well-designed and decisive experiments. These results show that free polycationic chains with a length of more than ~10 nm are able to partially block the fusion between different endocytic vesicles, including the endocytic-vesicle-to-endolysosome pathway. This thesis is highly original and its results greatly deepen our understanding of polymer-mediated gene transfection. More importantly, it provides new insights into the rational design of next-generation superior polymeric gene-delivery vectors.

From Nucleic Acids Sequences to Molecular Medicine by Volker A. Erdmann,Jan Barciszewski Pdf

Despite a half century of structural, biophysical and biochemical investigations of ribonucleic acids, they are still mysterious. RNAs stand at fertile crossroads of disciplines, integrating concepts from genomics, proteomics, dynamics as well as biochemistry and molecular biology. From 20 years it is clear, that genetic regulation of eukaryotic organisms has been misunderstood for the last years that the expression of genetic information is effected only by proteins. Basic understanding of nucleic acids has enhanced our foundation to probe novel biological functions. This is especially evident for RNA molecules whose functionality, maturation, and regulation require formation of correct secondary structure through encoded base-pairing interactions.

Spherical Nucleic Acids by Chad A. Mirkin Pdf

Spherical nucleic acids (SNAs) comprise a nanoparticle core and a densely packed and highly oriented nucleic acid shell, typically DNA or RNA. They have novel architecture-dependent properties that distinguish them from all other forms of nucleic acids and make them useful in materials synthesis, catalysis, diagnostics, therapeutics, and optics/plasmonics. This book covers over two decades of Dr. Mirkin’s research on SNAs and their anisotropic analogues, including synthesis and fundamental properties, and applications in colloidal crystallization, adaptive matter, and nanomedicine, spanning extra- and intracellular diagnostics, gene regulation, and immunomodulation. It is a reprint volume that compiles 101 key papers from high-impact journals in this research area published by the Mirkin Group at Northwestern University, Illinois, USA, within the International Institute for Nanotechnology, and collaborators. Volume 1 provides an overview and a historical framework of engineering matter from DNA-modified constructs and discusses the enabling features of nucleic acid–functionalized nanomaterials. Volume 2 covers design rules for colloidal crystallization, building blocks for crystal engineering, and DNA and RNA as programmable bonds. Volume 3 discusses colloidal crystallization processes and routes to hierarchical assembly, dynamic nanoparticle superlattices, surface-based and template-confined colloidal crystallization, optics and plasmonics with nanoparticle superlattices, and postsynthetic modification and catalysis with nanoparticle superlattices. Volume 4 covers diagnostic modalities, and intracellular therapeutic and diagnostic schemes based upon nucleic acid–functionalized nanomaterials.
