Many fungi and bacteria that associate with plants are potentially harmful and can cause disease, while others enter into mutually beneficial sym bioses. Co-evolution of plants with pathogenic and symbiotic microbes has lead to refined mechanisms of reciprocal recognition, defense and counter defense. Genes in both partners determine and regulate these mechanisms. A detailed understanding of these genes provides basic biological insights as well as a starting point for developing novel methods of crop protection against pathogens. This volume deals with defense-related genes of plants and their regulation as well as with the genes of microbes involved in their interaction with plants. Our discussion begins at the level of populations and addresses the complex interaction of plant and microbial genes in multigenic disease resistance and its significance for crop protection as compared to mono genic resistance (Chap. 1). Although monogenic disease resistance may have its problems in the practice of crop protection, it is appealing to the experimentalist: in the so-called gene-for-gene systems, single genes in the plant and in the pathogen specify the compatibility or incompatibility of an interaction providing an ideal experimental system for studying events at the molecular level (Chaps. 2 and 4). Good progress has been made in identifying viral, bacterial, and fungal genes important in virulence and host range (Chaps. 3-6). An important aspect of plant-microbe interactions is the exchange of chemical signals. Microbes can respond to chemical signals of plant origin.

Major advances in the areas under study have been reviewed in plenary lectures and are developed in the main articles of this book. Over 160 high-quality posters were presented and are summarized in short articles. Data from outstanding posters, which were discussed after a short oral presentation, are found in extended articles. As a whole the book presents a collection of papers arranged in six sections and reflecting the present day state-of-the-art of research in the field of plant defense reactions.

Much of that progress has been captured in the chapters of this book. Breakthroughs have been made possible through integration of molecular and whole-plant aspects. The classical boundaries between physiology, biochemistry and molecular biology have vanished. There has been a strong focus on a limited number of model species, including Arabidopsis thaliana. That focus has allowed greater insight into the significance of specific genes for plant development and functioning. However, many species are very different from A. thaliana, in that they are mycorrhizal, develop a symbiosis with N2-fixing microsymbionts, or have other specialised root structures. Also, some have a much greater capacity to resist extreme environments, such as soil acidity, salinity, flooding or heavy-metal toxicities, due to specific adaptations. Research on species other than A. thaliana is therefore pivotal, to develop new knowledge in plant sciences in a comprehensive manner. This fundamental new knowledge can be the basis for important applications in, e.g., agriculture and plant conservation. Although significant progress has been made, much remains to be learnt. It is envisaged that discoveries made in the recent past will likely lead to major breakthroughs in the next decade.

A NATO Advanced Study Institute on "Active Defence Mechanisms in Plants" was held at Cape Sounion, Greece, 21 April - 3 May 1981. It succeeded a similar Institute held at Porte Conte, Sardinia in 1975 on "Specificity in Plant Diseases. " What are active defence mechanisms in the context of plant disease in which a plant, the host, may be damaged by a pathogen? Defence mechanisms comprise properties of the host that decrease this damage. The mechanisms are passive when they are independent of the pathogen. They are active when they follow changes in the host caused by the pathogen. Thus for a fungal pathogen, cell walls of a higher plant which are lignified before infection would be a passive defence mechanism if they decreased damage by impeding growth of the fungus. Cell walls known to become lignified as a response to the pathogen would be an active defence mechanism if it were established that this response decreased damage. The papers and discussions at this Advanced Study Institute were about active defence mechanisms in higher plants, mainly econo mically important crop plants, against fungi, bacteria and viruses as pathogens. Taking the microorganisms first it is a truism but one that bears repeating that although plants almost always grow in close association with a wide variety of fungi and bacteria, often of types that can be pathogens, they rarely become diseased, at least not sufficiently so as to attract notice.
