Structural and Functional Repercussions of Loss of Function and Clinical Mutants of Presenilin by Omar Nyabi Pdf

Alzheimer's disease is a major health problem. The disease is clinically characterized by the progressive mental decline of the patients and pathologically by the accumulation of amyloid plaques and tangles and neurodegenerative changes in the brain. The cause of the disease remains unclear but in some cases, genetic (missense) mutations in the Amyloid Precursor Protein (APP) and in the Presenilin genes (PS1 & PS2) are sufficient to initiate the whole disease cascade. All these mutations cause in essence an increase in the generation or a change of the characteristics of the AB peptide making it more prone to produce amyloid plaques. The AB peptide is generated from the APP protein and the question how Presenilin influences APP processing and AB production is the main topic of our work.

Alzheimer’s Disease by J.D. Turner,K. Beyreuther,F. Theuring Pdf

Alzheimer's Disease is a progressive neurodegenerative disorder of late life with devastating consequences for the afflicted and their carers and poses one of the major challenges to medical research. Until recently, little hope of effective therapies capable of slowing the disease process or preventing its occurrence was apparent. With recent advances in the genetics and molecular biology of the disease processes and the demonstration of the involvement of multiple aetiological factors, however, real chances are now appearing for the identification of preventive drugs. In this discussion, experts from disciplines ranging from molecular genetics to the clinic provide review and novel data concerning the aetiology of AD and the establishment of drugfinding screening methods.

Cellular Peptidases in Immune Functions and Diseases 2 by Jürgen Langner,Siegfried Ansorge Pdf

Of the many special roles played by proteolytic enzymes in immune reactions, this book addresses different aspects of membrane peptidases, signal transduction via ligation of membrane peptidases (especially of dipeptidyl peptidase IV/CD26 and aminopeptidase N/CD13), and regulation of membrane peptidases in vivo and in vitro. A number of newly discovered peptidases (including cathepsin F, W and X, carboxypeptidase X, attractin) are described, with special emphasis given to the role of peptidases in immune and defense reactions and in the pathogenesis of inflammatory and other diseases, including rheumatoid arthritis, pancreatitis, multiple sclerosis, Alzheimer's disease and tumours of various origins. The focus on the involvement of a selection of proteolytic enzymes in immune reactions and diseases is a unique feature of this multifaceted work , which combines biochemical, immunological and clinical research reports with literary reviews of the field.

Alzheimer's Disease by Nigel M. Hooper Pdf

Alzheimer's disease is the most common cause of senile dementia. Since the discovery in 1984 of the amyloid ?-peptide (A?) as the core protein of the senile plaques present in the brains of Alzheimer's disease sufferers, an immense amount of research has gone into mapping out the molecular basis of this debilitating disease. The aim of Alzheimer's Disease: Methods and Protocols is to bring together the main biochemical, cell biological, and molecular biological techniques and approaches that are being used to investigate the molecular basis of Alzheimer's disease. This volume begins with chapters of an introductory/ review nature. Chapter 1 provides a historical introduction to Alzheimer's d- ease with particular emphasis on the central role played by A? and its re- tion to tau. Chapter 2 examines the genetics underlying this neurodegenerative disease, covering the amyloid precursor protein, apolipoprotein E, and the presenilins. Chapter 3 presents an overview of currently available therapeutic agents and prospects for drugs of the future.
