Human Papillomavirus and Cervical Cancer by International Agency for Research on Cancer Pdf

This volume reviews the evidence for a causal link between sexually transmitted infection with human papillomavirus (HPV) and the occurrence of cervical cancer, from a variety of different angles. Epidemiological studies and clinical, pathological, and cytological aspects of HPV infection are reviewed. Modern methods for analyzing HPV-DNA types by molecular biological techniques are described, and the statistical problems to be overcome in epidemiological work are explained. The volume was prepared by a broad team of experts from around the world, who met in Copenhagen in March 1988 to reach a consensus on the present state of understanding and to establish directions for future work.

Human Papillomavirus (HPV)-Associated Oropharyngeal Cancer by Daniel L. Miller,M. Sharon Stack Pdf

Head and neck squamous cell carcinoma (HNSCC), the sixth most prevalent cancer worldwide, remains a very difficult disease to treat and cure despite intensive investigation into molecular etiologies and tumor progression pathways. Due to public health efforts encouraging smoking cessation, the overall incidence of HNSCC has decreased in recent years in many countries. In contrast, the incidence of oropharyngeal squamous cell carcinoma (OPSCC) has increased significantly, and this subtype of HNSCC is commonly associated with human papillomavirus (HPV) infection. Moreover, individuals with HPV-positive OPSCC are generally younger and are frequently non-smokers, suggesting that HPV-associated OPSCC represents a distinct biologic entity. This volume summarizes the spectrum of current HPV-associated OPSCC research from the fundamental basic science to translational surgery and treatment approaches. Chapters are contributed by authoritative leaders in the fields of research and clinical care. Initial chapters address epidemiology, behavioral correlates of HPV infection, and racial disparities in oropharyngeal cancer. This is followed by chapters detailing HPV virology with focus on viral transformation, viral replication, and host response to viral infection. The molecular biology of HPV-associated OPSCC is investigated in chapters detailing alterations in signaling networks and unique mutational profiles of human tumors. Clinical presentation, surgical perspectives, and treatment paradigms specific to HPV-associated OPSCC conclude the volume. This comprehensive volume provides an up-to-date overview of both scientific discovery and clinical management of this emerging public health problem.

Molecular Determinants of Head and Neck Cancer by Barbara Burtness,Erica A. Golemis Pdf

Squamous cell cancers of the head and neck (SCCHN), also known as head and neck cancers (HNC) encompass malignancies of the oral cavity, larynx, nasopharynx and pharynx, and are diagnosed in over 500,000 patients worldwide each year, accounting for 5% of all malignancies. In the past several years, there have been significant developments in understanding of HNC. It is now recognized that although alcohol and tobacco use has represented the likely predominant cause of SCCHN, the incidence of a second class of SCCHN related to oncogenic human papillomavirus (HPV) infection is increasing, with a four-fold increase in the past 2 decades, and now thought to represent up to 30% of cases. The first effective target for SCCHN, the EGFR-targeting antibody cetuximab, was approved as recently as in 2006; since then, a growing body of research has identified additional signaling pathways as important in disease pathogenesis, and in resistance to treatment. Proteins such as c-Met, Src, and HER2 are emerging as new therapeutic targets, with a considerable ferment in the clinical trial community. As a capstone of research progress, 2011 marked the first reports of high throughput sequencing of SCCHN tumors, with these efforts identifying unexpected players such as Notch as frequent subject of mutation, spawning new hypotheses for future research. This book will be of interest to researchers who are interested in better understanding the biology of head and neck cancers, with the goals of better designing therapies, identifying risk factors, or investigating the molecular basis of the disease.

Advances in Radiotherapy for Head and Neck Cancer by Giuseppe Carlo Iorio,Nerina Denaro,Isacco Desideri,Umberto Ricardi,Valerio Nardone ,Lorenzo Livi Pdf

Modern Radiotherapy (RT) plays a key role in the management of Head and Neck Cancer (HNC). More precise delivery techniques, advanced image-guidance, and adaptive treatments characterize modern RT, enabling safer treatments with enhanced therapeutic window. Although patients identify the cure as their most important treatment outcome, complications related to treatment are a recognized problem as follow-up increases among those cured within this oncologic setting. This is particularly relevant for HPV-related oropharyngeal cancer (OPSCC), as these patients are younger, healthier, and more reactive to treatment. Thus, given the longer life expectancy, the jeopardizing impact of side effects on quality of life (QoL) and psychosocial functioning represent a forefront topic for HNC Researchers. De-escalation protocols have been developed recently, and, although not definitive, evidence is growing. This pertains particularly, but not exclusively, to HPV-related OPSCC.

Human Papillomavirus and Related Diseases by Davy Vanden Broeck Pdf

Cervical cancer is the second most prevalent cancer among women worldwide, and infection with Human Papilloma Virus (HPV) has been identified as the causal agent for this condition. The natural history of cervical cancer is characterized by slow disease progression, rendering the condition, in essence, preventable and even treatable when diagnosed in early stages. Pap smear and the recently introduced prophylactic vaccines are the most prominent prevention options, but despite the availability of these primary and secondary screening tools, the global burden of disease is unfortunately still very high. This book will focus on the clinical aspects of HPV and related disease, highlighting the latest developments in this field.

Human Papillomavirus and Head and Neck Cancer by Heather Walline Pdf

In the study of head and neck squamous cell carcinoma (HNSCC), we know that virally induced malignancies behave differently than those generated through other carcinogenic mechanisms. This paradigm extends into differences in tumor growth, treatment response and resistance, as well as invasion and metastasis. The inability to correctly diagnose and appropriately treat HNSCC at early stages has contributed to reduced survival for HNSCC patients, particularly those with HPV-negative disease. The examination of the molecular differences between HPV-positive and HPV-negative HNSCC will provide support for early diagnostic markers and guide appropriate treatment selection to improve patient care and survival. This Special Issue highlights current research efforts to understand the evolving contribution of human papillomavirus in HNSCC.

Betel-quid and Areca-nut Chewing and Some Areca-nut-derived Nitrosamines by IARC Working Group on the Evaluation of Carcinogenic Risks to Humans,International Agency for Research on Cancer Pdf

A working group of sixteen experts from seven countries re-evaluated the evidence of the carcinogenicity of betel-quid and areca-nut chewing and some areca-nut related nitrosamines. Betel-quid and areca-nut chewing are widely practised in many parts of Asia and in Asian-migrant communities elsewhere in the world. There are hundreds of millions of users worldwide. They evaluated betel quid with tobacco as carcinogenic to humans (Group 1) on the basis of sufficient evidence of an increased risk of cancer of the oral cavity, pharynx and oesophagus. The working group reviewed epidemiological studies of human cancer, mainly studies from India, Pakistan and Taiwan (China). Studies on betel quid with tobacco and areca nut with tobacco in experimental animals now also provide sufficient evidence of carcinogenicity. The working group also evaluated betel quid without tobacco as carcinogenic to humans (Group 1), on the basis of sufficient evidence of an increased risk of oral cancer. Studies on betel quid without tobacco and areca nut without tobacco in experimental animals now also provide sufficient evidence of carcinogenicity. Areca nut, a common ingredient of betel quid and many different chewing preparations, including those available commercially, has been observed to cause oral submucous fibrosis
