New Insights into Molecular Mechanisms Underlying Neurodegenerative Disorders by Chiara Villa,Yam Nath Paudel,Christina Piperi Pdf

Neurodegenerative disorders encompass a broad range of sporadic and/or familial debilitating conditions characterized by the progressive dysfunction and loss of selective neuronal populations, determining different clinical phenotypes. Emerging research data indicate an interplay of genetic factors and epigenetic mechanisms underlying neurodegenerative processes, which lead to increased prevalence of neurodegenerative disorders. In concert with the constant increase in the aging population, neurodegenerative disorders currently represent a major challenge to public health worldwide. Despite recent advances in clinical and preclinical research, the pathogenesis of these disorders still remains poorly understood, without effective treatments being available to halt the neurodegenerative processes, but rather aiming at relieving symptoms. Therefore, a critical evaluation of current research data and in-depth understanding of the molecular mechanisms that lead to neurodegeneration are crucial in order to identify potential therapeutic targets that can pave the way to the development of novel and promising therapies. This Special Issue is focused on novel molecular data in the field of neurodegeneration that associate with the onset and progression of neurodegenerative diseases. We are particularly interested in original articles and reviews that provide new insights into the main molecular pathogenic mechanisms underlying neurodegenerative disorders, aiming to identify potential biomarkers and novel therapeutic strategies.

The Prime Cause of Cancer by Otto Warburg,Trung Nguyen Pdf

This is book 2 of 5 of the “Understand Cancer” series. It is based on the best-available science. The SECONDARY causes of cancer were discussed in book one. This book continues from book one and discusses the PRIME cause of cancer as discovered by Nobel Prize Laureate Dr. Otto Warburg—considered by many as the founder of modern biochemistry. “There are prime and secondary causes of diseases. For example, the prime cause of the plague is the plague bacillus, but secondary causes of the plague are filth, rats, and the fleas that transfer the plague bacillus from rats to man. By a prime cause of a disease I mean one that is found in every case of the disease...Cancer, above all other diseases, has countless secondary causes. But, even for cancer, there is only one prime cause. Summarized in a few words, the prime cause of cancer is the replacement of the respiration of oxygen in normal body cells by a fermentation of sugar. All normal body cells meet their energy needs by respiration of oxygen, whereas cancer cells meet their energy needs in great part by fermentation. All normal body cells are thus obligate aerobes, whereas all cancer cells are partial anaerobes. From the standpoint of the physics and chemistry of life this difference between normal and cancer cells is so great that one can scarcely picture a greater difference. Oxygen gas, the donor of energy in plants and animals is dethroned in the cancer cells and replaced by an energy yielding reaction of the lowest living forms, namely, a fermentation of glucose.” —Dr. Otto Warburg

Sirtuin Biology in Medicine: Targeting New Avenues of Care in Development, Aging, and Disease provides a fascinating and in-depth analysis of sirtuins in the body during normal physiology as well during disease highlighting the targeting of sirtuin-controlled pathways for the development of innovative, efficacious, and safe therapeutic strategies for multiple disorders in the body that ultimately can affect lifespan extension. Sirtuins are expressed throughout the body, have broad biological effects, and can significantly impact both cellular survival and longevity during acute and long-term illnesses. These histone deacetylases play an intricate role in the pathology, progression, and treatment of several disease entities ranging from neurodegenerative disorders, cardiovascular disease, immune system dysfunction, reproductive dysfunction, endocrine disorders, gastrointestinal disease, drug dependency, and aging-related disorders. Implementing a translational medicine format, this unique reference highlights novel signaling pathways for sirtuins that promote stem cell proliferation, enhance cellular protection, modulate pathways of apoptosis and autophagy, and extend life span. Each chapter is presented with insightful detail that will be of interest and a comprehensive resource to audiences that include scientists, physicians, pharmaceutical industry experts, nutritionists, and students. Chapters are authored by internationally recognized experts who discuss the broad role of sirtuins in health and disease Details the basic and clinical role of sirtuins for the development of new clinical treatments Summarizes the multidiscipline views and publications for the compelling discipline of sirtuins by covering systems throughout the body Serves as an important resource for a broad audience of healthcare providers, scientists, drug developers, and students in both clinical and research settings

Pathobiology of Alzheimer's Disease by Anonim Pdf

Neuroscience Perspectives provides multidisciplinary reviews of topics in one of the most diverse and rapidly advancing fields in the life sciences. Whether you are a new recruit to neuroscience, or an established expert, look to this series for 'one-stop' sources of the historical, physiological, pharmacological, biochemical, molecular biological and therapeutic aspects of chosen research areas. The last decade has seen tremendous advances in our understanding of the pathobiology of Alzheimer's disease. These will lead to the first generation of drugs aimed at prevention rather than cure. This book covers some of the most important and exciting of these advances, with chapters written by many of the leading researchers in the field. With genetic studies as a backbone to this volume many chapters are devoted to the function and regulation of amyloid b-protein precursor (APP) and apolipoprotein E (ApoE). Other chapters describe cell biological approaches helping to piece together the link between the genetic alterations and the phenotype we call Alzheimer's disease. Although APP and its proteolytic cleavage product, amyloid b-protein, do not answer all the questions, detailed research into this system has undoubtedly increased our knowledge of the pathobiology of AD and has lead to the identification of other risk factors. Understanding the role of ApoE in the pathology of Alzheimer's disease promises to open a whole new field in AD research. * * Reviews the current knowledge of the pathogenesis of Alzheimer's Disease from a clinical perspective to a genetic and cell biological perspective * A comprehensive description of the role of amyloid B-protein precursor in Alzheimer's disease. * Up-to-date research data * Clear illustrations complement the text
