Production of Biologicals from Animal Cells in Culture reviews the state of the art in animal cell biotechnology, with emphasis on the sequence of events that occur when generating a biological from animal cells in culture. Methods that enable adjustment of nutrient feed streams into perfusion bioreactors so as to increase productivity are described. A number of issues are also addressed, such as the usefulness of the fingerprint method for cell characterization. Comprised of 135 chapters, this book begins with an overview of the problems and benefits of animal cell culture, followed by a discussion on the isolation of immortal murine macrophage cell lines. The reader is systematically introduced to the use of DNA fingerprinting to characterize cell banks; immortalization of cells with oncogenes; lipid metabolism of animal cells in culture; and energetics of glutaminolysis. Subsequent chapters explore serum-free and protein-free media; the physiology of animal cells; gene expression in animal cell systems; and animal cell bioreactors. The monitoring and assay of animal cell parameters are also considered, along with downstream processing and regulatory issues. This monograph will be of interest to students, practitioners, and investigators in the fields of microbiology and biotechnology.

Animal Cell Culture and Production of Biologicals by Ryuzo Sasaki,Kouji Ikura Pdf

In the past two decades, the importance of animal cell technology has increased enormously. First, useful proteins can be produced by cultured animal cells, in which the desired product can be modified and organized so as to retain its biological function. Second, studies of cultured cells can provide information needed to understand molecular mechanisms that govern what happens in tissues, organs, and even entire organisms. For this second purpose, biochemists and molecular biologists may need a large number of such cells. Third, cultured cells can be used instead of tissues and organs clinically. The Third Annual Meeting of the Japanese Association for Animal Cell Technology (JAACT), at which participants from abroad were warmly welcomed, was held in Kyoto on December 11-13, 1990. It was organized around the idea of providing a place for the review of much new data on such applications of cultured cells and for exchanges of the views of the participants about progress in the field. This volume, divided into seven sections, contains the proceedings of the meeting. The first section reviews the molecular basis of the control of animal cell growth. In the following sections, physicochemical and biochemical factors for cell growth and production of biologicals, cell culture systems including serum-free culture, new cell lines, specific products and their characteristics, and in vitro assays for toxic, carcinogenic, and pharmacological effects are taken up in their tum.

Animal Cell Culture by Mohamed Al-Rubeai Pdf

Animal cells are the preferred “cell factories” for the production of complex molecules and antibodies for use as prophylactics, therapeutics or diagnostics. Animal cells are required for the correct post-translational processing (including glycosylation) of biopharmaceutical protein products. They are used for the production of viral vectors for gene therapy. Major targets for this therapy include cancer, HIV, arthritis, cardiovascular and CNS diseases and cystic fibrosis. Animal cells are used as in vitro substrates in pharmacological and toxicological studies. This book is designed to serve as a comprehensive review of animal cell culture, covering the current status of both research and applications. For the student or R&D scientist or new researcher the protocols are central to the performance of cell culture work, yet a broad understanding is essential for translation of laboratory findings into the industrial production. Within the broad scope of the book, each topic is reviewed authoritatively by experts in the field to produce state-of-the-art collection of current research. A major reference volume on cell culture research and how it impacts on production of biopharmaceutical proteins worldwide, the book is essential reading for everyone working in cell culture and is a recommended volume for all biotechnology libraries.

Animal Cells as Bioreactors by Terence Cartwright Pdf

Because of their complexity, the new generation of genetically engineered protein drugs can only be made by biotechnological methods, using cultures of animal cells. This book covers all aspects of the technologies needed to turn animal cells into an acceptable and cost-effective tool for drug production. This includes modifying them genetically so that they produce the right product in high yield, getting them to grow reproducibly on an industrial scale, and extracting the required product from them. It also covers biological safety issues, and the verification of the chemical and biological nature of the protein drug produced. The work covers developments in all of these areas and how they all need to be integrated for the design of an effective biotechnological production process. It therefore provides a comprehensive guide to this area of biotechnology.

Fundamental and Applied Aspects of Animal Cell Cultivation by J. P. Barford Pdf

The advent of modern, biological techniques such as hybridoma technology, recombinant DNA techniques and viral transformation of cells has made the continuous production of a wide variety of biologicals possible using animal cells. The use of such products is well established in many diagnostic and (increasingly) therapeutic applications - the U.S. market for antibodies, for example, has been projected to increase from a 1991 level of US$0.33 billion to 1998 level of US$3.8 billion. Total sales of such products in 1992 was US$4.2 billion. The increasing application of this technology depends on increasing the efficiency of production and bioseparation and addressing various safety issues. This book examines the fundamental and applied aspects of animal cell cultivation.

Animal Cell Biotechnology by Raymond Spier,R. E. Spier,J. B. Griffiths Pdf

The Animal Cell Biotechnology series provides an in-depth state-of-the art coverage of a field which has developed into a multi-billion dollar industry over the last ten years. Written by leading experts, each volume contains well-referenced, fully-indexed reviews on practical and theoretical aspects of a specific subject, including: Bioreactor design and development Cell products and their development Downstream processing Product testing The main theme of this volume - applications of animal cells - has been broadened from the recurring theme of production of biologicals to the cells themselves as a product, and cells as a pharmacological/toxicological assay system. The need to replace animals in research and for the testing of drugs, foods and cosmetics is becoming increasingly critical as society has succeeded in making this a high profile ethical issue. It is the perceived belief that the best chance of reducing the use of animals will be to develop suitable assays based on cells in culture. Two chapters in this volume cover replacement animal cells and the use of skin cells for in vitro testing. Other chapters also detail the cell products tPA, Factor VIII, and Growth Factors. The basic tool is, of course, the cell and the text contains chapters on how to standardize the cell and process in order to achieve reproducible conditions, whether for production or assay purposes. Also featured is a review of immortalization techniques that can be used to construct new cell lines which fulfill the specialized criteria required of them. Finally a chapter on surface active agents is included.

Cell Substrates by J. C. Petricciani Pdf

This volume stems from a symposium sponsored by the W. Alton Jones Cell Science Center, Lake Placid New York. The Second Annual W. Alton Jones Cell Science Center Symposium: Cell Substrates and Their Use in the Production of Vaccines and Other Biologicals was held October 23-26, 1978. The Center is an operational unit of the Tissue Culture Association and offers, in collaboration with the Association's Education Committee, a wide range of educational and research activities. During the past 20 years there have been numerous national and international conferences on the topic of cell cultures used to produce biological products. Those largely dealt with the technology and associated issues that were current at the time of the meetings. For example, as human diploid cells were developed and proposed for use in vaccine production, a number of meetings were held to examine the pros and cons of human diploid cells. A large amount of data was provided at those conferences which formed the basis for the eventual acceptance of that cell system. Each meeting added to the general base of knowledge in the area of cell cultures and their application to the current and novel set of problems encountered. In general, the participants reaffirmed the basic premises that were formulated in the early days of polio virus vaccine production regarding the criteria for acceptability of cells when used in the manufacture of biologics intended for humans.
