Cajal and contemporary scientists have laid the basis of the modem concepts of the organization of the nervous system: the cir cuits of the brain are made up of individual neurons which transfer information via specialized structures called synapses. Soma and den drites usually receive the inputs, then the signal is carried all along the axon to the target areas. To fulfIll this task several types of neurons have developed their unique geometry characterized by a large recep tive area (soma and dendrites) and an often very extensive distal branching with the axon terminals. The volume of cytoplasm which constitutes the neuronal periphery is often far larger than the cell body, where the synthetic machinery is located. It is one of the roles ofaxoplasmic transport to supply the periphery with proper material and to sustain the specialized structures necessary for the physiological activity of the neuron. Furthermore, it has become more and more clear that target areas also exert effects on the innervating neurons, and these effects are not only mediated via recurrent fibers. Synapses have been shown to be able to pick up material from the synaptic left which is then intra axon ally transported back to the cell body. This retrograde axoplasmic transport has therefore been recognized as another basic mechanism to convey signals from the periphery to the centre.

The proceedings of a workshop conference are presented in this volume entitled Hypothalamic Peptide Hormones and Pituitary Regulation. The workshop was held in Wilson Hall on the campus of the National Institutes of Health, Bethesda, Maryland, during the days of November 1-2, 1976, and is the most recent of three symposia on neuroendocrinology that have been sponsored by the National Institutes of Health. The first one was held on December 6 - 8, 1961, in the New Everglades Hotel at Miami, Florida. During the first meeting, much emphasis was given to the anatomical and physiological basis for the fledgling science of neuroendocrinology. The proceedings of that symposium were published under the title of Advances in Neuroendocrinology, A. V. Nalbandov (ed. ), University of Illinois Press, Urbana, Illinois, 1963. The second workshop was held on January 8 -11, 1969, in the Arizona Inn at Tucson, Arizona, and was unique in several respects. It was evident to the participants that definitive identification and the determination of the chemical structure of at least one hypothalamic releasing factor was at hand (see Workshop Conference on Bioassay and Chemistry of the Hypophysio tropic Hormones of the Hypothalamus: ~Critical Evaluatioi'i':-J. Meites, ed. , The Williams and Wilkins Co. , Baltimore, Maryland, 1970). Much of what was presented at the second workshop was dedicated to methods of bioassay of the various releasing factors.

The present monograph is an imaginative and courageous attempt to provide a synthesis of knowledge concerning the af ferent connections of the medial basal hypothalamus. Only somebody who has lived through most of the explosive develop ment - over the last 25 years or so - in the neuroscience in general, and in hypothalamic functional neuroanatomy in parti cular, can fully appreciate the remarkably consistent picture emerging from this study. The writer of this foreword was (alas!) an active participant in the very early and premature, and also largely naive, attempts to penetrate the "jungle" of hypothalamic connections with degeneration methods when they first became available in the late 1930s. (I have told a part of this rather pathetic story is an autobiographical sketch in Pioneers in Neuroendocrinology [Meites et aI., (eds) (1975), Vol I. Plenum] because I was sufficiently self-critical not to publish my early results.) Even with the suppressive Nauta-type silver stains, introduced in the mid-1950s, studies of hypothala mic connections had only marginal results, which the reader will certainly appreciate if he compares the relevant figures in the first edition of Hypothalamic Control of the Anterior Pituitary [Szentagothai et al. (1962) Akademiai Kiad6, Budapest], with Dr. Zaborszky's concluding diagrams. The approach used by Dr. Zaborszky of combining the more advanced Fink-Heimer type degeneration techniques, and some of their most recent modifications by Gallyas et al.

It is only relatively recently that neurochemists and neurobiologists have shown appreciable interest in the class of macromolecules now generally re ferred to as complex carbohydrates, although gangliosides were, of course, first identified and studied in brain. The glycosaminoglycans fell chiefly within the province of connective-tissue biochemists, and earlier informa tion concerning the structure and metabolism of glycoproteins was largely limited to the more accessible glycoproteins and oligosaccharides (such as those found in plasma, milk, and urine), or ones which are relatively simple to prepare in a soluble and manageable form. Techniques were later devised for the isolation and purification of tightly bound membrane glycoproteins, where initial studies concentrated mainly on the erythrocyte, for which large amounts of a single cell population are available. Because of the structural complexity of nervous tissue and the large numbers, low concentrations, and membrane-bound form of many of its complex carbohydrates, progress has occurred more slowly in this area.

The "functional" in the title of this book not only reflects my personal bias about neuroanatomy in brain research, it is also the gist of many chapters which describe sophisticated ways to resolve structures and interpret them as dynamic entities. Examples are: the visualization of functionally identified brain areas or neurons by activity staining or intracellular dye-iontophoresis; the resolution of synaptic connections between physiologically identified nerve cells; and the biochemical identification of specific neurons (their peptides and transmitters) by histo- and immunocytochemistry. I personally view the nervous system as an organ whose parts, continuously exchanging messages, arrive at their decisions by the cooperative phenome non of consensus and debate. This view is, admittedly, based on my own ex perience of looking at myriads of nerve cells and their connections rather than studying animal behaviour or theorizing. Numerous structural studies have demonstrated that interneurons in the brain must receive hundreds of thousands of synapses. Many neurons receive inputs from several different sensory areas: each input conveys a message about the external world and possibly also about past events which are stored within the central nervous system. Whether an interneuron responds to a certain combination of inputs may be, literally, a matter of debate whose outcome is decided at the post synaptic membrane. A nerve cell responding to an overriding command is possibly a rare event.
