This exceptional laboratory manual describes thirty-seven procedures most likely to be used in the next decade for molecular, biochemical, and cellular studies on Drosophila. They were selected after extensive consultation with the research community and rigorously edited for clarity, uniformity, and conciseness. The methods included permit investigation of chromosomes, cell biology, molecular biology, genomes, biochemistry, and development. Each protocol includes the basic information needed by novices, with sufficient detail to be valuable to experienced investigators. Each method is carefully introduced and illustrated with figures, tables, illustrations, and examples of the data obtainable. The book's appendices include key aspects of Drosophila biology, essential solutions, buffers, and recipes. An evolution of Michael Ashburner's 1989 classic Drosophila: A Laboratory Manual, this book is an essential addition to the personal library of Drosophila investigators and an incomparable resource for other research groups with goals likely to require fly-based technical approaches.

This second edition volume expands on the previous edition by presenting updated protocols for several of the techniques described in the first edition of Drosophila: Methods and Protocols and current methods that cover recent breakthroughs in Drosophila research. The book begins with a description of FlyBase--a database of genes and genomes--followed by the presentation of systems for versatile gene expression in the fly. The first few chapters in this book detail gene knockdown and editing, including CRISPR-Cas9 and protein knockdown. The next few chapters are devoted to methods describing live imaging of different tissues and organs, followed by chapters on how to quantify image data and how to probe tissue mechanics by laser ablation. The next two chapters provide methods for analyzing transcription followed by protocols to study growth, metabolism, ageing, and behavior in Drosophila. This volume concludes with chapters on electrophysiological recordings and methods to establish cell lines. Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Cutting-edge and thorough, Drosophila: Methods and Protocols, Second Edition is a valuable source of hands-on protocols and reviews for molecular, cell, and developmental biologists using Drosophila as model systems in their work.

Apoptosis Methods and Protocols by Hugh J. M. Brady Pdf

The most fundamental question facing each and every cell within an org- ism is to survive or to die. Cell death is required for normal function; some estimates suggest that as many as one million cells undergo cell death every second in the adult human body. Almost all cells undergoing physiological, or programmed, cell death, independent of cell type, manifest a stereotypic p- tern of morphological changes termed apoptosis. Typically, apoptotic cells d- play shrinkage, membrane blebbing, chromatin condensation, and nuclear fragmentation. The integrity of the cell membrane is not lost during apoptosis and so avoids eliciting the inflammatory response that would have been caused by the spillage of the cell’s contents. This is quite in contrast to the loss of cell contents typical of necrosis. The caspases, the family of intracellular cysteine proteases associated with apoptosis, are responsible for the stereotypical m- phological changes. Caspases cleave various substrate proteins that act on DNA fragmentation, nuclear envelope integrity, the cytoskeleton, and cell volume regulation. Apoptotic cells are cleared in vivo by the process of phagocytosis, in which specific “phagocytes” move to the site of apoptosis, engulf the dying cells and digest them. Apoptosis has a central role in many physiological processes, for example, in the immune system. Autoreactive cells are deleted via apoptosis to prevent autoimmunity. At the end of an immune response, activated lymphocytes are removed to maintain homeostasis within the immune system.

Fundamental Approaches to Screen Abnormalities in Drosophila by Monalisa Mishra Pdf

This volume includes in-depth, hands-on protocols for detecting various developmental defects in Drosophila. It provides cutting-edge methods for maintaining Drosophila under laboratory conditions to perform various experiments, and for dissecting various imaginal discs of the larvae. Further, biochemical protocols for estimating the levels of different metabolites, reactive oxygen species, and fat-sensitive pathways are discussed, and various staining and behavior techniques for fat detection are provided. The book explains how various fluorescent dyes and the comet assay can be used to identify DNA damage, and elaborates on the analysis of the eye, antennae, imaginal disc, gut and muscle damage under bright field and fluorescent microscopes. It covers the analysis of hemolymph using Giemsa staining; determining the functionality of channel proteins, eye and mechanosensory organs in adults and larvae using various behavioral assays; and metal detection and structural analysis for various tissues using SEM. In closing, it addresses the analysis of Drosophila’s eye and head via paraffin section; measurement of reactive oxygen species from various tissues via FACS; and the CRISPER technique for gene editing and analysis of micro RNA mutations.
