Systems Biology of MicroRNAs in Cancer by Ulf Schmitz,Olaf Wolkenhauer,Julio Vera-González Pdf

This book provides an update on the latest development in the field of microRNAs in cancer research with an emphasis on translational research. Since the early 2000s, microRNAs have been recognized as important and ubiquitous regulators of gene expression. Soon it became evident that their deregulation can cause human diseases including cancer. This book focuses on the emerging opportunities for the application of microRNA research in clinical practice. In this context, computer models are presented that can help to identify novel biomarkers, e.g. in circulating microRNAs, and tools that can help to design microRNA-based therapeutic interventions. Other chapters evaluate the role of microRNAs in immunotherapy, immune responses and drug resistance. Covering key topics on microRNAs in cancer research this book is a valuable resource for both emerging and established microRNA researchers who want to explore the potential of microRNAs as therapeutic targets or co-adjuvants in cancer therapies.

Systems Biology and the Challenge of Deciphering the Metabolic Mechanisms Underlying Cancer by Osbaldo Resendis-Antonio,Christian Diener Pdf

Since the discovery of the Warburg effect in the 1920s cancer has been tightly associated with the genetic and metabolic state of the cell. One of the hallmarks of cancer is the alteration of the cellular metabolism in order to promote proliferation and undermine cellular defense mechanisms such as apoptosis or detection by the immune system. However, the strategies by which this is achieved in different cancers and sometimes even in different patients of the same cancer is very heterogeneous, which hinders the design of general treatment options. Recently, there has been an ongoing effort to study this phenomenon on a genomic scale in order to understand the causality underlying the disease. Hence, current “omics” technologies have contributed to identify and monitor different biological pieces at different biological levels, such as genes, proteins or metabolites. These technological capacities have provided us with vast amounts of clinical data where a single patient may often give rise to various tissue samples, each of them being characterized in detail by genomescale data on the sequence, expression, proteome and metabolome level. Data with such detail poses the imminent problem of extracting meaningful interpretations and translating them into specific treatment options. To this purpose, Systems Biology provides a set of promising computational tools in order to decipher the mechanisms driving a healthy cell’s metabolism into a cancerous one. However, this enterprise requires bridging the gap between large data resources, mathematical analysis and modeling specifically designed to work with the available data. This is by no means trivial and requires high levels of communication and adaptation between the experimental and theoretical side of research.

An investigation of microRNA target regulation mechanisms using an integrative approach by Ulf Schmitz Pdf

This work is a showcase for the integration of systems biology and bioinformatics tools, algorithms and models for deciphering biological phenomena. More specifically, it integrates (i) prediction algorithms for identifying and characterizing molecular interactions, (ii) structural modelling of molecule complexes, (iii) network analysis approaches, and (iv) mathematical modelling and simulation. Two comprehensive workflows are implemented for the analysis of collective target gene regulation by microRNAs and for the prediction of cooperating microRNA pairs and their mutual target genes. In two case studies mechanisms of fine-tuned target gene regulation are revealed for different cellular processes and the phenomenon of cooperative target regulation is identified as frequent mechanism of gene regulation in humans.

MicroRNA as Biomarkers in Cancer Diagnostics and Therapy by Lorenzo F. Sempere Pdf

This Special Issue celebrates the 25th anniversary of the discovery of the first microRNA. The size of the microRNome and complexity of animal body plans and organ systems suggests a role for microRNAs in cell fate determination and differentiation. More than 2000 sequences have been proposed to represent unique microRNA genes in humans, with an increasing number of mechanistic roles identified in developmental, physiological, and pathological processes. Thus, dysregulation of a few key microRNAs can have a profound global effect on the gene expression and molecular programs of a cell. This great potential for clinical intervention has captured the interest and imagination of researchers in many fields. However, very few fields have been as prolific as the field of cancer research. This Special Issue provides but a glimpse of the large body of literature of microRNA biology in cancer research, containing 4 original research studies and 4 review articles that focus on specific hematologic or solid tumors in disease. Collectively, these articles highlight state-of-the-art approaches and methodologies for microRNA detection in tissue, blood, and other body fluids in a range of biomarkers applications, from early cancer detection to prognosis and treatment response. The articles also address some of the challenges regarding clinical implementation.
